Title RNA-seq analysis of high fat diet-induced changes in macrophage transcriptome

Summary Systemic low-grade inflammation associated with obesity and the metabolic syndrome is also a strong risk factor for the development of diabetes. This inflammatory state is primarily caused by the release of proinflammatory cytokines by macrophages. However, the effect of obesity on the macrophage transcriptome, including long non-coding RNAs, is not known. Here, we analyzed global changes in gene expression of elicited peritoneal macrophages from mice fed a high-fat diet (HFD) or control diet for 12 weeks. The HFD-fed mice has significantly increased body weight and impaired glucose tolerance compared to controls. Transcriptome analysis showed that HFD incuced the expression of genes associated with inflammatory response.
 
Overall design C57BL/6J mice were fed with standard chow or high-fat diet (HFD, 60%kcal, Research Diets Inc., D12492i) for 12 weeks. Mice were administered with 3% thioglycollate medium via intraperitoneal injection, and four days later, peritoneal macrophages were collected.
