Summary |
Cosmc is an obligate chaperone for C1GalT1 T transferase necessary for generating mature mucin-type O-glycans. Animals lacking Cosmc in glomerular podocytes develop proteinuria, glomerulosclerosis and progressive renal failure. This study was designed to examine transcriptome differences in 1 month male animals with and without podocyte Cosmc, a time point when proteinuria is established but glomerular architecture is still relatively intact.

Overall design |
Glomeruli were isolated by Dynabead perfusion technique from 1 month old control(n=5) and Podo-Cre; CosmcY/flox(n=4) male animals. RNA was isolation using Rneasy Plus Mini kit (Qiagen). Samples were sent to GeneWiz for polyA-based mRNA enrichment, cDNA synthesis, RNA-seq library preparation and sequencing on an Illumina HiSeq. Paired end 2x150 bp reads were obtained. Bioinformatic analysis was performed through GeneWiz
