Summary |
In the present study, a distinct transcriptome regulated by NO2-CLA was revealed in primary human coronary artery endothelial cells (HCAECs) through RNA sequencing. Differential gene expression and pathway enrichment analysis identified numerous regulatory networks of cell cycle regulation, inflammation and redox responses, nitric oxide biosynthesis, hypoxia and chaperones in response to NO2-CLA, suggesting potent and diverse effects of NO2-CLA on cellular processes.

Overall design |
For RNA-seq experiment, HCAECs at 90% confluence were treated with DMSO, CLA (10µM) or NO2-CLA (10µM) for 1 hour, followed by adding with either ethanol (Control) or PA (200 µM) (Cayman Chemical) for the subsequent 6 hours. There are 6 groups for RNA seq: DMSO + control (ethanol), CLA (10µM) + control, NO2-CLA (10µM) + control, DMSO + PA, CLA (10µM) + PA, NO2-CLA (10µM) + PA. Each group has 4 biological replicates.
