Summary |
Mesenchymal stem cells (MSCs) are involved in tissue homeostasis and repair, and can be impaired in subjects with high fat diet (HFD)-induced metabolic disorders. We used microarrays to detail the global programme of gene expression bone marrow MSCs (BMSCs) underlying HFD-induced progression of metabolic disorders in rat and identified distinct classes of differentiatilly expressed genes during this process.

Overall design |
Six-week old Sprague-Dawley (SD) rats were randomized to normal diet (ND) and high-fat diet (HFD) groups, fed a normal diet (ND, 100% basal diet) and a customized high-fat diet (60% basal diet, 10% sucrose, 20% lard and 10% egg yolk powder), respectively. The total mRNA of BMSCs (P1) from both groups (n= 3 per group) at the specified feeding time points (1, 2 and 4 months) was purified and hybridized on Affymetrix microarrays. BMSCs were isolated and characterized for the expression of cell surface markers before RNA extraction, to improve the accuracy of expression profiles.
