Summary |
Purpose: The goals of this study are to use RNA-seq to identify genes whose expression are regulated by protein ISGylation in a genome-wide scale Methods: mRNA profiles of PyVmT/Uba7 KO + Control and PyVmT/Uba7 KO+UBE1L mammary epithelial cells (MECs) were generated by deep-sequecing in duplicates, using Illumina HiSeq4000. Results: Using an optimized data analysis workflow, we mapped about 25 million sequence reads per sample to the mouse genome (M16 genome annotation). 259 genes are identified as differentially expressed transcripts from RNA-seq analysis of PyVmT/Uba7 KO + empty vector vs PyVmT/Uba7 KO + UBE1L MECs treated by LPS (100ng/mL) for 6 hours after IFN (1000 U/mL) priming for 24 hours. Conclusions: protein ISGylation facilitated expression of a group genes, which are regulated by STAT1/STAT2 containing complexes.
