Summary The goal of this study is to analyze the role of Srf in transcriptional analysis during cardiomyocyte maturation in mouse
 
Overall design Srf-F/F (mutant) and Srf-F/+ (control) mice were injected with AAV9-cTNT-Cre to deplete Srf specifically in cardiomyocytes in vivo at two different stages. Three biological replicates were analyzed in each group. Thus a total of 12 samples were analyzed.
In neonatal knockout model, AAV was injected at post natal day 1 (P1) and cardiomyocytes collected at P14. In adult knockout model, AAV was injected at P60 and cardiomyocytes collected at P90
In each experiment, cardiomyocytes were isolated by retrograde collagenase perfusion and purified by fluorescence activated cell sorting.
Total RNA was extracted from FACS sorted cells. RNA-seq libraries were generated using SMART-seq and Nextera kits and were sequenced on a NextSeq500 sequencer. FASTQ Data were aligned by STAR and count matrix generated by FeatureCounts.
