Summary |
In this study, we conducted a genome-wide screen to identify differentially expressed long noncoding RNAs (lncRNAs), circular RNAs (circRNAs), micro RNAs (miRNAs) and mRNAs during replication and transcription using a tetracistronic transcription and replication-competent virus-like particle (trVLP) system that models the life cycle of EBOV in 293T cells.

Overall design |
P1 trVLPs were used to infect 293T cells and the TCID50 of P1 trVLPs was 10^-6.5/ml. Approximately 4×10^5 cells were present in each well of a 6-well plate, and P1 trVLPs used for infection in this experiment were applied in a volume of 2 ml per well. There were three wells per time point (0 hour, 24 hour, 48 hour, 72 hour and 96 hour) for the sequencing step. The samples in each well was used for independent experiments.
