Deciphering m6A dynamics at single-base level during planarian anterior-posterior (AP) axis specification

(Submitter supplied) Background: The establishment of the anterior-posterior (A-P) axis is a crucial step during tissue repair and regeneration. Despite the association reported recently of N6-methyladenosine (m6A) with regeneration, the mechanism underlying the regulation of m6A in A-P axis specification during regeneration remains unknown. Herein, we deciphered the m6A landscape at a single-base resolution at multiple time points during A-P axis regeneration and constructed the de novo transcriptome assembly of the Dugesia japonica planarian.

ACME dissociation: a versatile cell fixation-dissociation method for single-cell transcriptomics

(Submitter supplied) Single-cell sequencing technologies are revolutionizing biology, but are limited by the need of dissociating fresh samples that can only be fixed at later stages. We present ACME (ACetic-MEthanol) dissociation, a species-versatile cell dissociation approach that fixes cells  as they are being dissociated. ACME-dissociated cells have high RNA integrity, can be cryopreserved multiple times, can be sorted by Fluorescence-Activated Cell Sorting (FACS) and are permeable, enabling combinatorial approaches of single-cell transcriptomics. more...

Regenerative adaptation to electrochemical perturbation in planaria: a molecular analysis of physiological plasticity

(Submitter supplied) The development and maintenance of organismal form results from dynamic interactions between genome, protein interaction, physiology and the external environment. Bioelectric signaling represents an important epigenetic layer of control over processes of large-scale patterning during development and regeneration. However it is still largely unknown how physiological plasticity and adaptation operate during regenerative processes. more...

Deep Sequencing identifies regulated small RNAs in Dugesia japonica

(Submitter supplied) We applied Illumina sequencing to identify microRNAs (miRNAs) and piwi-interacting small RNAs (piRNAs) in Dugesia japonica. Dugesia were cut up after seven day’s starvation. DJ1 is the whole Dugesia body, DJ2 is the head part, DJ3 is the tail part, DJ4 is the left part and DJ5 is the right part. Total RNA was extracted by Trizol, and preserved in ethanol, stored at -80°C until further use.

Planarian stem cells

(Submitter supplied) Difficulty of experimental accessing makes in vivo stem cell studies not easy in mammals and model systems with regenerative capability represent an alternative for the characterization of stem cell-specific genes. Planarians retain a population of pluripotent stem cells, the neoblasts, throughout their adult life. Neoblasts can be selectively destroyed by X-ray allowing us to compare stem cell-less organisms with wild-type worms. more...
