Scanning iron response regulator binding sites using RNA-seq and Dap-seq in the Brucella genome

(Submitter supplied) Brucella spp. are gram-negative, facultative intracellular pathogens that cause brucellosis in humans and animals. Iron is an essential element required for all organisms. Iron response regulator (Irr) is a crucial transcriptional regulator and can affect the growth and iron uptake of Brucella. The growth rate of Brucella melitensis M5-90 irr mutant was significantly lower than that of B. melitensis M5-90 under normal or iron-sufficient conditions, however, the growth rate of the B. more...

Temporal Gene Expression Analysis of the intracellular pathogen Brucella melitensis following bovine Peyer's patch infection.

(Submitter supplied) In a time course study, we characterized global gene expression profile of B. melitensis infection in bovine Peyer patches in the first 4 h p.i. Statistical analysis of microarray results reveal that 2,356 B. melitensis genes were detected as differentially expressed (z-score p < 0.025) compared with control between 15 min and 4 h post-infection. A group of 1,740 genes that were differentially expressed in at least 4 out of 5 time points was considered the core set of genes that reflect the major changes in B. more...

Global Rsh-dependent transcription profile of Brucella suis during stringent response unravels adaptation to nutrient starvation and cross-talk with other stress responses.

(Submitter supplied) In the intracellular pathogen Brucella spp., the activation of the stringent response, a global regulatory network providing rapid adaptation to a variety of growth-affecting stress conditions such as nutrient deficiency, is essential for replication in the host. A single, bi-functional enzyme Rsh catalyzes synthesis and hydrolysis of the alarmone (p)ppGpp, responsible for differential gene expression under stringent conditions. more...

It has been shown to be involved in a wide range of cellular processes and pathways. To characterize the regulation roles of Hfq in Brucella melitensis, the in vitro hfq induction conditions simulating intracellular environments were firstly defined, and then, the transcriptome of the hfq mutant was compared to that of the wild type strain. more...

Intracellular Brucella melitensis gene expression

(Submitter supplied) Brucella spp. is an intracellular pathogen in vivo. The intracellular B. melitensis transcriptome was determined by initially enriched and then amplified B. melitensis RNA from total RNA of B. melitensis-infected HeLa cells. Analysis of microarray results identified 161 and 115 genes differentially expressed at 4 and 12 h p.i., respectively. Most of the genes (78%) differentially expressed were down-regulated at the earliest time point, but up-regulated (75%) at 12 h p.i. more...

Differential Expression of Iron Acquisition Genes by Brucella melitensis and Brucella canis During Macrophage Infection (part 1)

(Submitter supplied) Brucella melitensis and Brucella canis differ by ~75 genes yet B. melitensis is highly virulent for humans while B. canis is considered rarely pathogenic. No identified bacterial factors or mechanisms account for this difference in virulence. To identify functional differences of these two bacteria, gene transcription was examined during infection of murine macrophages and compared to bacteria grown in broth. more...

Comparative transcriptome analysis of 16M and its vjbR mutant under a condition simulating environments of macrophage

(Submitter supplied) Quorum sensing system (QS) is a global regulator that regulates gene expression in response to cell density and other environmental factors. Two QS genes have been identified in Brucella, one of which (vjbR) is involved in Brucella intracellular survival and virulence. But the regulation mechanism of Brucella intracellular survival by the vjbR remains unknown. To characterize the regulation roles of vjbR, the in vitro vjbR induction conditions simulating intracellular environments were firstly defined, and then, transcriptome of the vjbR mutant is compared to that of the wild type strain. more...

Identification of MucR regulatory targets

(Submitter supplied) The transcriptional regulator MucR was identified as highly attenuated in a mutagenesis screen conducted by this laboratory. MucR has been described as a key regulator of exopolysaccharide synthesis in closely related Rhizobials soil symbionts. The goal of this study was to apply custom Brucella melitensis oligonucleotide microarrays to identify the regulatory targets of MucR. These studies revealed that MucR's regulation of exopolysaccharide synthesis is also conserved in Brucella melitensis and is also a key regulator of iron sequestering/storage and denitrification. more...

Expt 2: The contribution of vjbR and N-dodecanoyl homoserine lactone on gene regulation in Brucella melitensis

(Submitter supplied) Quorum sensing (QS) is a communication system in bacteria that regulates gene expression in response to a small diffusible signal indicative of the bacterial population present. QS has been shown to regulate virulence genes, biofilm formation, cell division and secretion systems in a diverse set of bacteria. Brucella melitensis produces a QS signaling molecule, N-dodecanoyl homoserine lactone (C12-HSL), that interacts with a LuxR transcriptional regulator, VjbR. more...

Quorum Sensing regulation in Brucella melitensis 16M

(Submitter supplied) Many pathogenic bacteria use a regulatory process termed Quorum Sensing (QS) to produce and detect small diffusible molecules to synchronize gene expression within a population. In Gram-negative bacteria, the detection and response to these molecules depend on transcriptional regulators belonging to the LuxR family. Such a system have been discovered in the intracellular pathogen Brucella melitensis, a Gram-negative bacteria responsible for brucellosis, a word-wide zoonosis remaining a serious public health concern in endemic countries. more...

Brucella melitensis global gene expression

(Submitter supplied) In this study we report that B. melitensis at the late logarithmic phase of growth are more invasive for HeLa cells than at mid logarithmic or stationary growth phases. Microarray analysis of B. melitensis gene expression identified 414 up- and 40 down-regulated genes in late-log growth phase compared to the stationary growth phase. The vast majority of the up-regulated genes in late-log cultures were those associated with DNA replication, transcription and translation, intermediate metabolism, energy production and conversion, membrane transport and cell envelope, biogenesis and outer membrane, while the down-regulated genes were distributed among several functional categories. more...
