Acarbose improves obesity-induced insulin resistance and immune homeostasis by directly regulating gut microbiota and macrophages inflammation

(Submitter supplied) Acarbose was reported to alleviate obesity-induced insulin resistance IR and regulate the gut microbiota community; however, the critical microbial signaling metabolites and the host targets associated with the metabolic benefits of acarbose remains unknown. In this study, we reported that acarbose treatment was able to significantly improve high fat diet (HFD) induced IR and inflammation states in adipose tissue and intestine by influencing the survival and pro-inflammatory function of M1 macrophages. more...

Single-cell Analysis Reveals the Protective Role of Anxa1-expressing Macrophages Induced by Bifidobacterium Feeding in Acute Lung Injury Mouse Models

(Submitter supplied) Acute lung injury/acute respiratory distress syndrome (ALI/ARDS) is a life-threatening condition characterized by lung inflammation and damage. Mechanical ventilation can exacerbate this condition. The gut microbiome, known to impact health, might have implications for ALI/ARDS outcomes. This study aimed to investigate the effects of probiotics in a murine ALI model. Using a two-hit approach combining lipopolysaccharide-induced inflammation and mechanical ventilation-induced injury, a severe lung injury model was established in mice. more...

Inhibition of adipose triglyceride lipase alleviates non-alcoholic steatohepatitis in diabetic mice via impaired PPARa signalling favouring hydrophilic bile acid composition

(Submitter supplied) Inhibition of adipocyte triglyceride lipase (ATGL/PNPLA2) activity in insulin resistance and non-alcoholic fatty liver disease is an attractive therapeutic target. This study evaluated the impact of Atglistatin-mediated ATGL inhibition on non-alcoholic steatohepatitis (NASH) development in diabetic and hyperlipidaemic mice. Streptozotocin-injected male mice were fed an HFD to induce NASH. We analysed liver histology, hepatic lipid content, immunohistochemistry, RNA sequencing, and serum biochemistry. more...

Dynamic enhancer interactome promotes senescence and aging [Hi-ChIP II]

(Submitter supplied) Gene expression programs are regulated by enhancers which act in a context-specific manner, and can reside at great distances from their target genes. Extensive three-dimensional (3D) genome reorganization occurs in senescence, but how enhancer interactomes are reconfigured during this process is just beginning to be understood. Here we generated high-resolution contact maps of active enhancers and their target genes, assessed chromatin accessibility, and established one-dimensional maps of various histone modifications and transcription factors to comprehensively understand the regulation of enhancer configuration during senescence. more...

High-fat diet during mouse pregnancy impairs fetal heart development

(Submitter supplied) Maternal obesity is associated with detrimental outcomes in offspring. However, the specific effects of gestational exposure to a high-fat diet (HFD) on fetal heart development remain poorly understood. In the study, we investigated that maternal overnutrition accounted for defective development in offspring, and cell proliferation was restricted severely in neonatal organ. Starting with this clue, we found decreased cell cycle and similar cross area of cardiomyocytes in offspring exposed to a maternal HFD. more...

Comparison of the gene expression profile in aorta tissue between C57BL/6J and Apoe-knockout mouse

(Submitter supplied) The physiological functions other than lipid metabolism of APOE may interfere with the research results of Apoe-knockout mice as atherosclerosis models. The elucidation of the molecular basis of the physiological functions of APOE other than lipid metabolism will help to make rational use of the Apoe-knockout mouse model. Therefore, our aim is to reveal the changes in aortic gene expression profile caused by Apoe-knockout. more...

Enhanced metabolic resilience and exercise adaptation of skeletal muscle in males by Esr1-induced remodeling of mitochondrial cristae-nucleoid architecture

(Submitter supplied) Reduced estrogen action is associated with obesity and insulin resistance. However, the cell and tissue-specific actions of estradiol in maintaining metabolic health remain inadequately understood especially in men. We observed that skeletal muscle ESR1/Esr1 (encodes estrogen receptor a), including expression of specific ESR1 variants is positively correlated with insulin sensitivity and metabolic health in humans and mice. more...

Effects of Allulose and Erythritol Consumption on a High-Fat Diet

(Submitter supplied) Long-term consumption of erythritol, a widely used sugar substitute, has been associated with increased risks of thrombosis and cardiometabolic diseases. In this study, we investigated the effects and mechanisms of allulose in mitigating these risks compared to erythritol using the clusterProfiler tool. Since a high-fat diet (HFD) is known to enhance platelet aggregation, we compared the pathways related to these processes between groups of mice treated with allulose and those treated with erythritol. more...

Fasting reshapes tissue-specific niches to improve the innate immune response to cancer

(Submitter supplied) Fasting has long been associated with the improved elimination of cancer cells. However, the distinctive role of specific immune subsets mediating these anti-tumor properties remain elusive. Using a cyclic fasting diet (CFD) after tumor initiation, we investigated the impact of fasting on NK cell anti-tumor immunity. We found that adherence to a CFD improved anti-tumor immunity against both solid and metastatic tumors in a NK cell-dependent manner. more...

Hepatic Gene Expression Induced by CREB-H-ΔTC

(Submitter supplied) The cAMP-response element binding protein H (CREB-H) is a transcription factor expressed in liver and intestinal tissues. It is well known for regulating many lipid and glucose metabolism genes. Our group aimed to screen for additional or potential genes that may be downstream of CREB-H transcription. To prevent inaccuracy arisen from endogenous CREB-H expression, we used CREB-H-knockout (CREB-H KO) mice for this study.

Decreased mRNA expression of malonyl-CoA decarboxylase emerges from an inborn error-like defect in the Mlycd gene of DBA/2J mice

(Submitter supplied) Mlycd encodes malonyl-CoA decarboxylase (MCD), which is an enzyme that localizes in the cytosolic, mitochondrial, and peroxisomal compartments and catalyzes the conversion of malonyl-CoA into acetyl-CoA. Malonyl-CoA can be converted into malonylcarnitine (C3DC). Patients with an autosomal recessive defect of MCD and MCD KO mice have pronounced elevations of C3DC. Analysis of plasma C3DC levels in the BxD genetic reference population revealed increased levels in BxD strains that harbor the DBA/2J haplotype at the site of the Mlycd gene. more...
