RANKL effect on osteoclastogenic cell line

Analysis of H10 osteoclastogenic cells following treatment with soluble recombinant receptor activator of nuclear factor-kB ligand to induce osteoclastogenesis. H10 cells derived from RAW264.7 macrophages. Results provide insight into the molecular mechanisms regulating osteoclastogenesis.

CD109 plays a role in osteoclastogenesis.

(Submitter supplied) The primary aim of this project was to identify novel factors, in particular the cell-surface protein CD109, which regulate osteoclastogenesis. Microarray analysis was performed comparing two pre-osteoclast cell lines generated from the RAW 264.7 osteoclast cell line: one that has the capacity to fuse forming large multinucleated cells and one that does not fuse. It was found that CD109 was up-regulated by > 17-fold in the osteoclast forming cell line when compared to the cell line that does not fuse.

Transcriptional profiles of genes in the early stage of osteoclastogenesis

(Submitter supplied) Osteoclast (OC) differentiation undergoes a two-step process: commitment of hematopoietic progenitor cells to tartrate-resistant acid phosphatase (TRAcP) positive OC precursors (OCPs), and fusion of OCPs into multinucleated OCs. (Submitter supplied) Comparison of gene expression of the osteoclast precursor myeloid blast seeded on plastic and on bone, primed with M-CSF for 4 days and culture with M-CSF and RANKL for 1 day. Analysis of the role of c-Fos and Nfat in osteoclastogenesis by expression profiling of osteoclastogenic splenocytes lacking c-Fos but with an active Nfatc4. Active Nfatc4 introduced into cells by retroviral gene transfer. Osteoclastogenesis restored by active Nfatc4 in splenocytes lacking c-Fos.
