Pristimerin effect on myeloma cell lines

Analysis of H929 and U266 myeloma cells treated with the natural triterpenoid pristimerin. Multiple myeloma tumors exhibit dysregulated cyclin D expression. Pristimerin identified from a chemical library screen for compounds that induce suppression of transcription from the cyclin D2 promoter.

Expression data from myeloma cell lines treated with 500 nM pristimerin or DMSO vehicle alone for 4 hours

(Submitter supplied) As multiple myeloma tumors universally dysregulate cyclin D genes we conducted high-throughput chemical library screens for compounds that induce suppression of cyclin D2. The top-ranked compound was a natural triterpenoid, pristimerin. We used gene expression microarray studies to identify co-regulated pristimerin-response genes and to deduce the compound’s direct molecular target(s), utilizing pattern-matching algorithms available at the Connectivity Map (Cmap). more...

RNA sequencing of MM.1S and U266 cell lines after IT848 treatment

(Submitter supplied) IT848 is a small molecule inhibiting DNA binding of all five NF-kB subunits. In vitro treatment of lymphoma or multiple myeloma cells with IT848 suppresses NF-kB transcriptional activity, cell growth, and indices apoptosis.

A novel chordoma xenograft allows in vivo drug testing and reveals the importance of NF-kB signaling in chordoma biology [SNP array]

(Submitter supplied) Chordoma is a rare primary bone malignancy that arises in the skull base, spine and sacrum and originates from remnants of the notochord. These tumors are typically resistant to conventional chemotherapy, and to date there are no FDA-approved agents to treat chordoma. The lack of in vivo models of chordoma has impeded the development of new therapies for this tumor. Primary tumor from a sacral chordoma was xenografted into NOD/SCID/IL-2R γ-null mice. more...
