Gene Set Enrichment Analysis reveals lineage relationships between mouse and human gliomas and identifies stem cells and PNET as distinct entities

(Submitter supplied) Murine and human genome-wide microarray expression profiles (including GSE19404) were compared using Partek and Gene Set Enrichment Analysis (GSEA) in order to identify similarities. A murine PNET-like tumour by morphology was found to resemble human ATRT, whereas experimental glioma resembled a particular human glioblastoma subclass.

Genome-wide molecular characterisation of CNS PNET and Pineoblastoma

(Submitter supplied) sPNETs are highly malignant embryonal brain tumours of poor prognosis. The underlying biology is poorly understood. To address this we therefore performed high resolution genetic analysis. 36 CNS PNETs and 8 PBs were analysed using the Affymetrix 100K and 500K Mapping Set to identify copy number imbalance at both the chromosome and gene level. Keywords: Affymetrix 100K SNP array, Affymetrix 500K SNP arrays

Cooperative actions of p53 and Pten in normal and neoplastic progenitor cell renewal and differentiation

(Submitter supplied) Glioblastoma (GBM) is a highly lethal brain tumor presenting as one of two subtypes with distinct clinical histories and molecular profiles. The primary GBM subtype presents acutely as high-grade disease that typically harbors EGFR, PTEN and Ink4a/Arf mutations, and the secondary GBM subtype evolves from the slow progression of low-grade disease that classically possesses PDGF and p53 events1. Here, we show that concomitant CNS-specific deletion of p53 and Pten in the mouse CNS generates a penetrant acute-onset high-grade malignant glioma phenotype with striking clinical, pathological and molecular resemblance to primary GBM in humans. more...

Molecular subclasses of high-grade glioma: prognosis, disease progression, and neurogenesis

(Submitter supplied) Novel prognostic subclasses of high-grade astrocytoma are identified and discovered to resemble stages in neurogenesis. One tumor class displaying neuronal lineage markers shows longer survival, while two tumor classes enriched for neural stem cell markers display equally short survival. Poor prognosis subclasses exhibit either markers of proliferation or of angiogenesis and mesenchyme. Analysis of gene expression data is described in Phillips et al., Cancer Cell, 2006. more...

Vemurafenib resistance selects for highly malignant brain and lung-metastasizing melanoma cells

(Submitter supplied) V600E being the most common mutation in BRAF, leads to constitutive activation of the MAPK signaling pathway. The majority of V600E BRAF positive melanoma patients treated with the BRAF inhibitor vemurafenib showed initial good clinical responses but relapsed due to acquired resistance to the drug. The aim of the present study was to identify possible biomarkers associated with the emergence of drug resistant melanoma cells. more...

ADAMTSL2, a missing link in Wnt/ß-catenin regulated CNS vascular development

(Submitter supplied) Context dependent molecular cues shape the formation of the cerebral vascular network and the function of the blood-brain barrier (BBB). The Wnt/ß-catenin pathway is orchestrating CNS vascular development, but downstream mediators have not been characterized. Here we generated an endothelial cell-specific R26-Axin1 overexpression (AOE) mouse model to inhibit Wnt/ß-catenin signaling. In AOE mice we discovered that blockade of Wnt/ß-catenin pathway leads to premature regression and remodeling without compromising BBB integrity. more...
