Transcription factor Klf5 deficiency effect on embryonic stem cells

Analysis of embryonic stem cells (ESCs) lacking the transcription factor Klf5. Klf5 deficiency results in the failure of ESC derivation from cells of the inner cell mass and early embryonic lethality. Results provide insight into the role of Klf5 in blastocyst development and ESC self-renewal.

WT vs Klf5 KO ES

(Submitter supplied) Homozygous disruption of Bteb2/Klf5, a homolog of Drosophila gap gene Krüppel, led to increased expression of various differentiation marker genes, such as Fgf5, Cdx2, and Brachyury in mouse ES cells without compromising their ability to differentiate into all three germ layers. Upon removal of LIF, Klf5-deficient ES cells showed faster differentiation kinetics than wild-type ES cells. Genome-wide maps of O-GlcNAc-Oct4 binding regions in ZHBTc4 F-Oct4 mouse embryonic stem cells

(Submitter supplied) We report that Oct4 is modified by O-GlcNAc in stem cells. To find O-GlcNAc-Oct4 target genes, we ChIPed Oct4 with Flag(Oct4) antibody and then O-GlcNAc modified proteins are enriched by sWGA beads (succinylated wheat germ agglutinin (sWGA), which specifically binds O-GlcNAc). Results show that several genes implicated in pluripotency regulation are bound by O-GlcNAc-Oct4 in their gene region.

Metastable primordial germ cell-like state induced from mouse embryonic stem cells by Akt activation
