LPS-induced and E. coli-induced gene expression in wildtype and MyD88-/- mouse macrophages

(Submitter supplied) Comparison of gene expression in wildtype and MyD88-/- C57BL/6J mouse macrophages treated with 10 ng/mL LPS for 2 hours versus media treated control macrophages, and, wildtype and MyD88-/- C57BL/6J mouse macrophages treated with live E. coli bacteria (log phase; 1 bact per 1 macrophage) for 2 hours versus media treated control macrophages. Cells from 4 mice of each geneotype were used and each individual provided its own control. more...

MyD88-deficient macrophage response to lipopolysaccharide and E. coli (dye swap)

Analysis of MyD88 null mutant macrophages treated with LPS or live E. coli. MyD88 transduces cell signaling events downstream of Toll-like receptors, a key component of host defense. Results suggest most of the host response to endotoxin or live bacteria is actually regulated independently of MyD88.

Microarray analysis of activated mixed glial (microglia) and monocyte-derived macrophage gene expression.

(Submitter supplied) Since macrophage activation can now be studied at a global level using modern microarray and proteomic analyses, discovery of novel macrophage activation genes is inevitable and important for understanding HIV-associated dementia (HAD). We isolated two different types of primary human macrophages, microglia and monocyte-derived macrophages (MDM) from brain tissue and whole blood respectively. The microarray analysis of differentially regulated macrophage activation genes reported here supports our previous assertions that the mixed glia (MIX) cultured in starvation conditions (DMEM alone) are a non-activated, or “quiescent”, tissue culture model for studying macrophage activation in the brain. more...

MPSS profiling of embryonic gonad and primordial germ cells

(Submitter supplied) We used the MPSS technology to uncover gene expression profiling in a greater depth in the early embryonic gonads and primordial germ cells (PGCs) in the chicken. Total numbers of sequenced signatures were 1,012,533 and 995,676 for the PGCs and gonad. Using a false discovery rate cut-off of 0.05, we found 3.05 % of all signatures in the PCGs and 1.89 % of all signatures in the gonad signatures were significantly up-regulated compared to each sample. more...

Gene expression profiles of adult peripheral and cord blood mononuclear cells altered by lipopolysaccharide

(Submitter supplied) Neonatal gram-negative sepsis is often characterized by a fulminat clinical course compared to adults resulting in higher morbidity and mortality. Genome-wide gene expression analysis can provide insights into the molecular alterations in sepsis. To evaluate in vitro activation of the neonatal and adult immune system, gene expression patterns were compared in mononuclear cells from cord (CBMNC) and adult peripheral blood (APBMNC). more...

Gene expression profiles of RAW264.7 macrophage cells treated with Paeony root-derived compounds

(Submitter supplied) Paeony root has long been used for its anti-inflammatory effects. In this study, the effects of albiflorin, paeoniflorin, and paeonol, compounds from paeony root, on gene expression profiles were examined in macrophages. The RAW264.7 macrophages were treated with LPS in the presence or absence of albiflorin, paeoniflorin, or paeonol. Global mRNA expression levels were detected by using an oligonucleotide microarray platform covering the mouse whole genome. more...

Comparison of antigen stimulation response in Mycobacterium bovis-infected vs. control cattle using BOTL-5 microarray

(Submitter supplied) Bovine tuberculosis (BTB) caused by Mycobacterium bovis continues to cause substantial losses to global agriculture and has significant repercussions for human health. The advent of high throughput genomics has facilitated large scale gene expression analyses that present a novel opportunity for revealing the molecular mechanisms underlying chronic mycobacterial infection. Using this approach, we have previously shown that innate immune genes in peripheral blood mononuclear cells (PBMC) from BTB-infected animals are repressed in vivo in the absence of exogenous stimulation. more...
