2,3,7,8-tetrachlorodibenzo-p-dioxin effect on cultured embryonic teeth

Analysis of cultured molar teeth from embryonic day 14 fetuses following in vitro treatment with 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD), a potent toxic dioxin. Results provide insight into the molecular basis of the retardation of dentin mineralization caused by TCDD.

E14 mouse molar tooth germs cultured with/without TCDD

(Submitter supplied) Dioxins are ubiquitous environmental poisons that are developmentally toxic. Exposure of mouse E14 tooth germs to 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) leads to reduced tooth size and deformation of cuspal morphology implying the induction of a variety of biological responses on both cellular and molecular levels. To verify such responses at the gene level, mouse embryonic cap staged tooth germs were cultured for 24 h with/without 1 µM TCDD. more...

Fetal mouse heart, TCDD dose-response series

(Submitter supplied) Pregnant C57Bl6N mice were treated with 0 (corn oil), 1.5, 3.0, or 6.0 ug/kg TCDD on gd14.5. Fetal hearts were collected on gd17.5. Hearts from each litter were pooled onto one chip. 4 replicates of each condition were run on affymetrix MG_U74Av2 chips, using standard affymetrix protocols and controls. Keywords: dose-response, 3 doses plus corn oil control, 4 replicates

2,3,7,8-tetrachlorodibenzo-p-dioxin effect on cardiovascular development

Analysis of gestational day (GD) 17.5 fetal hearts isolated from pregnant C57BL/6Ns treated at GD 14.5 with up to 6.0 ug/kg 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD). TCDD, a cardiovascular teratogen, inhibits cardiomyocyte proliferation, a process that peaks between GD 14.5 to 17.5.

Patterns of dioxin-altered mRNA expression in livers of dioxin-sensitive versus dioxin-resistant rats

(Submitter supplied) The dioxin congener 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) causes a wide range of toxic effects in rodent species, all of which are mediated by a ligand-dependent transcription-factor, the aryl hydrocarbon receptor (AHR). The Han/Wistar (Kuopio) (H/W) strain shows exceptional resistance to many TCDD-induced toxicities; the LD50 of >9600 µg/kg for H/W rats is higher than for any other wild-type mammal known. more...

NanoString analysis of a rat liver mRNA panel following TCDD treatment

(Submitter supplied) Profiling of mRNA abundances with high-throughput platforms such as microarrays and RNA-Seq has become an important tool in both basic and biomedical research. However these platforms remain prone to systematic errors, and have challenges in clinical and industrial application. As a result it is standard practice to validate a subset of key results using alternate technologies. Similarly, clinical and industrial applications typically involve transitions from high-throughput discovery platform to medium-throughput validation ones. more...

Identification of novel genes expressed during mouse molar tooth development by microarray gene expression analysis

(Submitter supplied) To identify genes heretofore undiscovered as critical players in the biogenesis of teeth, we have used microarray gene expression analysis of the developing mouse molar tooth (DMT) between 1 and 10 days postnatal to identify genes differentially expressed when compared to 16 control tissues (GEO accession # GSE1986). Of the top 100 genes exhibiting increased expression in the DMT, 29 were found to have been previously associated with tooth development. more...
