Ecdysone treatment and gamma-irradiation effect

Analysis of Kc167 cells following gamma-irradiation, ecdysone treatment, or both. Pretreatment of Kc167 cells with ecdysone increases the sensitivity of these cells to gamma-irradiation.

Ecdysone- and stress-induced expression profile in Kc167 cells

(Submitter supplied) Kc167 cells were mock-treated/treated with combinations of steroid hormone ecdysone and gamma-irradiation, and harvested. The expression profiles were determined using Affymetrix Drosophila Genechip 1 arrays. Comparisons between the sample groups allow the identification of genes with ecdysone- and/or radiation-responsive expression patterns. Keywords: response to hormone, stress response

Radiation-induced Dmp53-dependent expression profile

(Submitter supplied) Embryos were collected, aged, mock-treated/treated with 40Gy gamma radiation, and allowed to recover for 1.5 hours. Targets from 3 sets of wild type (yw, w1118) and 2 sets of mutant (Dmp53NS) biological replicates were generated and the expression profiles were determined using Affymetrix Drosophila Genechip 1 arrays. Comparisons between the sample groups allow the identification of genes with radiation-responsive and Dmp53-dependent expression patterns. more...

p53 mutant embryo response to radiation

Analysis of p53 mutant embryos exposed to gamma radiation. Within 90 minutes of radiation exposure, apoptosis is observed in embryonic and post-embryonic tissues of wild type but not p53 mutants. Results provide insight into the molecular basis of this p53-dependent apoptogenic response.

Characterization of the Drosophila genome at the nuclear lamina

(Submitter supplied) The nuclear lamina (NL) binds chromatin in vitro and is thought to function in its organisation, but genes that interact with the NL are unknown. Using an in vivo approach we identified 474 Drosophila genes that interact with B–type lamin, Lam. These genes are transcriptionally silent, late replicating, lack active histone marks, and are widely spaced. These factors collectively predict Lamin binding behavior, indicating the NL integrates variant and invariant chromatin features. more...

Expression data for whole adult tissue; Drosophila males of seven species

(Submitter supplied) Co-expression of genes that physically cluster together is a common characteristic of eukaryotic transcriptomes. Identifying these groups of co-expressed genes is important to the functional annotation of genomes and understanding the evolutionary fates of the clustered genes. We used microarrays to measure gene expression in seven closely related Drosophila species, to identify domains clusters within a species of Drosophila (D. more...

p53 status and BaP or BPDE gene expression response

(Submitter supplied) Human colon carcinoma cells (HCT116) differing in p53 status were exposed to benzo(a)pyrene (BaP) (2.5 and 5 uM for up to 48 h) or anti-benzo(a)pyrene-trans-7,8-dihydrodiol-9,10-epoxide (BPDE)(0.5 and 1 uM for up to 24 h), and their gene expression responses compared by cDNA microarray technology. Keywords: BaP or BPDE exposure

Coordinated regulation of cell cycle transcripts by p53-inducible microRNAs, miR -192 and -215

(Submitter supplied) Cell cycle arrest in response to DNA damage is an important anti-tumorigenic mechanism. microRNAs (miRNAs) were shown recently to play key regulatory roles in cell cycle progression. For example, miR-34a is induced in response to p53 activation and mediates G1 arrest by down-regulating multiple cell cycle-related transcripts. Here we show that genotoxic stress promotes the p53-dependent up-regulation of the homologous miRNAs, miR -192 and miR-215. more...
