Single-cell transcriptomics unveils xylem cell development and evolution

(Submitter supplied) The plant xylem tissue is responsible for water transportation, mechanical support and lateral growth, and can be utilized as wood. The seed plant xylem has a radial system composed of ray parenchyma cells and an axial system of fusiform cells, including tracheids in gymnosperms and vessel elements and libriform fibers in angiosperms. Little is known about the developmental programs and evolutionary relationships of these xylem cell types. more...

RNA-seq of major tissues and xylem cell types of Populus trichocarpa

(Submitter supplied) We used the stem cross-sections of P. trichocarpa and our recently developed laser capture microdissection (LCM) to collect fibers, vessels, and a combination of 3 cell types (fibers + vessels + rays). Total RNA from the three samples was isolated, amplified, and analyzed by full-transcriptome RNA-sequencing. The sequencing reads were mapped to the P. trichocarpa genome.

Overexpression of PtaHB1, Homeodomaine-leucine-Zipper class III, in hybrid poplar

(Submitter supplied) Transcriptional profiling of primary xylem tranformed cells comparing control empty vector to PtaHB-1 overexpressed Over-expression constructs were obtained by inserting the complete coding sequences of PtaHB1 and PtaHB7 from P. tremula x P. alba between the maize ubiquitin promoter (Christensen et al., 1992) and a 35S terminator into the pCambia1305.2 vector (www.cambia.org). The resulting plasmids were then transferred into A. more...

Genome-wide mapping of histone H3 lysine 4 trimethylation in Eucalyptus grandis developing xylem

(Submitter supplied) Histone modifications play an integral role in plant development, but have been poorly studied in woody plants. Investigating chromatin organization in wood-forming tissue and its role in regulating gene expression allows us to understand the mechanisms underlying cellular differentiation during xylogenesis (wood formation) and identify novel functional regions in plant genomes. However, woody tissue poses unique challenges for using high-throughput chromatin immunoprecipitation (ChIP) techniques for studying genome-wide histone modifications in vivo. more...

Transcriptome profiling of Pinus radiata branches reveals new insights into compression wood formation in response to gravity stress

(Submitter supplied) Compression (CW) and opposite wood (OW) are formed in the uniderside and upperside of conifer branches respectively in response to gravity stress. We investigated genes differentially transcribed between the underside and upside of radiate pine branches using cDNA microarrays with a view to plant gravitropism.

Arborea spruce 11K cDNA microarray

(Submitter supplied) The 11K microarray was designed based on the white spruce (Picea gluca [Moench.] Voss) expressed sequence tag (EST) assembly. This assembly comprised 16600 unique contigs sequenced from 17 cDNA libraries, which represented different tissues at a diverse developmental stages and growing conditions. 10400 nonredundant sequences were selected and printed as double, side-by-side on the microarray slide with 4X12 subgrid layout and 132 spots per subgrid. more...

PCAN_TRAUMATEST

(Submitter supplied) Gene expression analysis in response to mechanical injuries in Pinus canariensis, known for a great healing ability. Traumatic xylem formed in the margins of the wounds were compared to control tissues, sampled from remote parts of the plant, in order to check transcriptomical variations in three dates during the response: H1, H2 and H3 for 7, 78 and 92 days after wounding.
