Expression profiling in dopaminergic brain structures of rats self-administering cocaine

(Submitter supplied) Gene expression profiling in dopaminergic brain structures of rats self-administering cocaine. Effect of histone deacetylase inhibition We have shown that injection of the HDAC inhibitor trichostatin A (TsA) to rats is sufficient to decrease their motivation to self-administer cocaine. The aim of the present study was to investigate alterations in gene expression patterns in the Anterior Cingulate Cortex and Nucleus Accumbens of rats self-administering cocaine and treated repeatedly with TsA, and compare them with rats taking only cocaine. more...

CHD7 controls cerebellar development via Reelin

(Submitter supplied) Mutation of the gene encoding the ATP-dependent chromatin remodeler CHD7 causes CHARGE syndrome. The mechanisms underlying the neurodevelopmental deficits associated with the syndrome, which include cerebellar hypoplasia, developmental delay, coordination problems and autistic features, are not known. CHD7 is expressed in neural stem and progenitor cells, but its role in neurogenesis during brain development remains unknown. more...

RAS signalling through PI3-Kinase controls cell migration via modulation of Reelin expression

(Submitter supplied) RAS signalling through Phosphoinositide 3-kinase (PI3-Kinase) has been shown to have an essential role in tumour initiation and maintenance. RAS also regulates cell motility and tumor invasiveness, but the role of direct RAS binding to PI3-Kinase in this remains uncertain. Here, we provide evidence that disruption of RAS interaction with PI3-Kinase p110adecreases cell motility and prevents activation of Rac GTPase. more...

Genome-wide identification and characterization of functional neuronal activity-dependent enhancers

(Submitter supplied) Experience-dependent gene transcription is required for nervous system development and function. However, the DNA regulatory elements that control this program of gene expression are not well defined. Here we characterize the enhancers that function across the genome to mediate activity-dependent transcription in neurons. While ~12,000 putative activity-regulated enhancer sequences have previously been identified that are enriched for H3K4me1 and the histone acetyltransferase CBP, we find that this chromatin signature is not sufficient to distinguish which of these regulatory sequences are actively engaged in promoting activity-dependent transcription. more...

Gene expression data from cultured mouse cortical (mCTX) neurons in different stimulation and knockdown conditions

(Submitter supplied) Although the induction of C-FOS in the brain has been extensively studied for several decades to date there has been no attempt to identify the targets of C-FOS at a genome wide level, and it was not known how many genes C-FOS activates in a given cell. To identify potential C-FOS target genes, we performed microarray analysis on RNA obtained from mouse cortical (mCTX) neurons infected with lentivirus containing either a control shRNA (targeting firefly luciferase) or c-Fos shRNA that were subsequently depolarized with 0, 1, 3, or 6 hours of KCl.
