Role of calcium/calmodulin-dependent kinase type IV in podocyte function in lupus nephritis

(Submitter supplied) Kidney podocytes and their slit diaphragms contribute to prevent urinary protein loss. T cell from patients with systemic lupus erythematosus display increased expression of calcium/calmodulin kinase IV (CaMKIV). Here we evaluated the functional role of CaMKIV in lupus nephritis (LN) using kidney biopsy specimens and human podocyte cell line (AB8/13). We found that exposure of podocytes to IgG from LN patients resulted in entry of IgG into the cytoplasm. more...

Podocytes use FcRn to clear IgG from the glomerular basement membrane

(Submitter supplied) The glomerular filtration barrier prevents large serum proteins from being lost into the urine. It is not known, however, why the filter does not routinely clog with large proteins that enter the glomerular basement membrane (GBM). Here we provide evidence that an active transport mechanism exists to remove immunoglobulins that accumulate at the filtration barrier. We found that FcRn, an IgG and albumin transport receptor, is expressed in podocytes and functions to internalize IgG from the GBM. more...

CRAC channel controls the differentiation of pathogenic B cells in lupus nephritis

(Submitter supplied) Systemic lupus erythematous (SLE) is a prototype of autoimmune disease. Lupus nephritis (LN) is one of the most serious complications of SLE. The development of autoreactive B cells and the production of autoantibodies have been critical for the initiation and progression of LN. Store-operated Ca2+ entry (SOCE) is the main Ca2+ influx pathway in lymphocytes and is essential for immune response . SOCE is mediated by Ca2+ release-activated Ca2+ (CRAC) channels which are comprised of stromal interaction molecule (STIM) and calcium release-activated calcium modulators (ORAI) . more...

Loss of SIRPα promotes podocytes presenting antigen to activate specific T cell immune responses in lupus nephritis via activating spleen tyrosine kinase

(Submitter supplied) To investigate the role of SIRPα in podocytes, we generated podocyte-specific SIRPα knockout mice, Subsequent studies have found that SIRPa is deeply involved in the adaptive immune process of podocytes, and the loss of SIRPa promotes the ability of podocytes to present antigens, furthermore, the loss of SIRPa in podocytes can promote the increase of antigen presentation ability of podocytes and promote the progression of lupus nephrin

Maternal IgG Affects Brain Development of Mouse Pups

(Submitter supplied) Maternal IgG is an immunoglobulin present in breast milk and has been shown to play a role in the development of the immune system in infants. The aim of this study was to investigate the effects of maternal IgG on brain development in mouse pups. Maternal IgG immunoreactivity was observed in microglial cells of the pup brain, peaking at postnatal 8 day. In particular, strong IgG immunoreactivity was observed in the microglia in the corpus callosum and cerebellar white matter. more...
