Gene expression in TFH cells

(Submitter supplied) After activation, CD4+ helper T (Th) cells differentiate into distinct effector subsets. Although chemokine (C-X-C motif) receptor 5-expressing T follicular helper (Tfh) cells are important in humoral immunity, their developmental regulation is unclear. Here we show that Tfh cells had a distinct gene expression profile and developed in vivo independently of the Th1 or Th2 cell lineages. microRNA expression profiling of different T cell subset in human tonsils

(Submitter supplied) Tight control of follicular helper T(Tfh) cells is required for optimal maturation of the germinal center (GC) response. The molecular mechansims controlling Tfh cell differentiation remain incompletely understood. Here we sought to to identfiy miRNAs that might regulate Tfh cell development and/or function. To identfiy miRNAs that are differentially expressed in Tfh cells, we performed Agilent microRNA microarray analysis comparing Tfh cells with the other main T cell subsets (naive T cells, non-Tfh effector or non-Tfh memory T cells, CD57+ Tfh cells and CD57- Tfh cells).

Comparison of gene expression profile of T follicular helper cells and germinal center B cells from neonatal and adult mice

(Submitter supplied) Vaccines that require T-cell help for the production of antibodies need to be administered three to four doses during the first 15 months of life in order to afford protection. Diminished antibody responses in newborns have been considered as a consequence of impaired T follicular helper (TFH) development and germinal center B cell (GC BC) formation upon immunization. To gain further insight into the transcriptional program for the induction and/or inhibition of TFH cell development, we screened 561 immune-related genes using the nCounter® Immunology Panel array.

Gene expression of T follicular helper (TFH) cells 6 h after ICOS-L blockade

(Submitter supplied) The maintenance of the TFH phenotype depends on continuous signals via ICOS. For a global assessment of differences in gene expression after interruption of the ICOS pathway a genome wide transcriptome analysis was performed. We used the OT-II adoptive transfer system to isolate antigen-specific TFH cells (day 6 after immunization) after short-term (6 hours) blockade of the ICOS pathway using a monoclonal antibody against ICOS-L.

Gene array for identifying molecules involved in IL-10 regulation during Th17 polarization

(Submitter supplied) IL-10 production by Th17 cells is critical for limiting autoimmunity and inflammatory responses. Gene array analysis on Stat6 and T-bet double deficient Th17 cells identified the Th2 transcription factor c-Maf to be synergistically up-regulated by IL-6 plus TGFbeta, and associated with Th17 IL-10 production. Genome-wide analysis identifies Bcl6 repression of the IL-7R/STAT5 axis during T follicular helper (Tfh) cell differentiation

Lineage-specific functions of Bcl6  in immunity and inflammation are mediated through distinct biochemical mechanisms

(Submitter supplied) The transcription factor Bcl6 orchestrates the germinal center reaction through its actions in B and T cells, and regulates inflammatory signaling in macrophages. We report that genetic replacement by mutant Bcl6, which cannot bind corepressors to its BTB domain, disrupted  germinal center formation and immunoglobulin affinity maturation, due to a defect in B cell  proliferation and survival.
