Global changes in gene expression during compatible and incompatible interactions of cowpea (Vigna unguiculata L.) with the root parasitic angiosperm Striga gesnerioides

(Submitter supplied) Background. Cowpea, Vigna unguiculata L. Walp., is one of the most important food and forage legumes in the semi-arid tropics. While most cowpea accessions are susceptible to the root parasitic weed Striga gesnerioides, several cultivars have been identified that show race-specific resistance. Cowpea cultivar B301 contains the RSG3-301 gene for resistance to S. gesnerioides race SG3, but is susceptible to race SG4z. more...

Identification and analysis of microRNAs associated with drought tolerance in cowpea

(Submitter supplied) To identify novel microRNAs that are associated with drought tolerance in two different cowpea genotypes, we generated small RNA sequences from adult cowpea plants under control and dought stress treatments. Over 79 million raw reads were generated and numerous novel microRNAs are identified, including some associated with drought tolerance.

SSHscreen analysis of cowpea drought stress SSH libraries (F&R) to calculate Enrichment ratio 2 and 3 values

(Submitter supplied) Screening cDNA clones from SSH library by hybridization with cDNA used to construct the library. Treated samples were drought-stressed cowpea plants, and control samples were cowpea plants subjected to a standard watering regime. cDNA clones from forward and reverse libraries are spotted on the same array, but data from each library were analysed separately after normalization using SSHscreen software (http://microarray.up.ac.za/SSHscreen ) to calculate Enrichment Ratio 3 (ER3) and Enrichment Ratio 2 (ER2) values for each clone. more...

Rhizobial adaptation to hosts, a new facet in the legume root-nodule symbiosis

(Submitter supplied) Certain alpha- and beta-proteobacteria, the rhizobia, are able to infect legume roots, elicit root nodules, and live therein as endosymbiotic, nitrogen-fixing bacteroids. Host recognition and specificity are the results of consecutive programming events in bacteria and host plants in which important signaling molecules, e.g. plant flavonoids and rhizobial lipooligosaccharides, play key roles. Here, we introduce a new aspect of this symbiosis: the adaptive response to hosts. more...

Transcriptional profiling of root-knot nematode induced feeding sites in cowpea (Vigna unguiculata L. Walp)

(Submitter supplied) In this study we have looked at the transcriptome profile of both incompatible and compatible cowpea-RKN interaction for two different time points using the Affymetrix soybean GeneChip. This is the first study of this kind in cowpea-RKN interaction. This study provides a broad insight into the Rk-mediated resistance in cowpea and creates an excellent dataset of potential candidate genes involved in both nematode resistance and parasitism, which can be further tested for their role in this biological process using functional genomics approaches. more...

Detection and Validation of Single Feature Polymorphisms in Cowpea using a Soybean Genome Array

(Submitter supplied) Background: Cowpea (Vigna unguiculata L. Walp) is an important food and fodder legume of the semiarid tropics and subtropics worldwide, especially in sub-Saharan Africa. High density genetic linkage maps are needed for marker assisted breeding but are not available for cowpea. A single feature polymorphism (SFP) is a microarray-based marker which can be used for high throughput genotyping and high density mapping. more...

Cowpea_UP12_expression_array [110706_asp_bean_CB_expr]

(Submitter supplied) Each probe is replicated 3 times. Ten copies of each of eight control probes derived from the yeast genome sequence with no matches to the unigene sequences were also included in the microarray. A custom Roche NimbleGen microarray was designed based on 29,728 unigene sequences from assembly P12 of cowpea cDNAs in the HarvEST database (http://harvest.ucr.edu/). With the exception of 189 unigenes that failed to pass quality filtering for probe design, each of the unigenes was represented by a set of four 60-mer probes on the fabricated microarray. more...
