Comparative Genomic Hybridization (CGH) reveals sex chromosome location and gene movement in stalk-eyed flies (family Diopsidae), Teleopsis quinqueguttata

(Submitter supplied) We utilized Comparative Genomic Hybridization (CGH), using probes designed from de novo assembly of a testes transcriptome, to identify genes located on the sex chromosomes and autosomes of a stalk-eyed fly, Teleopsis quinqueguttata. Analysis of X chromosome gene content revealed the evolution of a neo-X chromosome that originated prior to the diversification of the family. Comparison of X-linkage across three species spanning the phylogenetic breadth of the family indicates abundant chromosomal gene movement, particularly for genes expressed exclusively in the testes.

Comparative Genomic Hybridization (CGH) reveals sex chromosome location and gene movement in stalk-eyed flies (family Diopsidae)

(Submitter supplied) We utilized Comparative Genomic Hybridization (CGH), using probes designed from de novo assembly of 10 tissues into a grand transcriptome to identify genes located on the X and Y chromosomes of a stalk-eyed fly, Teleopsis dalmanni. We use next generation sequencing of RNA to identify over 500 genes that are differentially expressed in the testes due to the effects of a driving X chromosome (XSR) in T. more...

Gene expression during head development in a sexually monomorphic stalk-eyed fly

(Submitter supplied) Stalk-eyed flies (family Diopsidae) are a model system for studying sexual selection due to the elongated and sexually dimorphic eye-stalks found in some species. These flies are of additional interest because their X chromosome is derived largely from an autosomal arm in other flies. To investigate how sex-biased expression arose on the novel X we compared gene expression between males and females using oligonucleotide microarrays and RNA from developing eyestalk tissue in the sexually monomorphic diopsid, Teleopsis quinqueguttata. more...

Sex-biased gene expression during head development in a sexually dimorphic stalk-eyed fly

(Submitter supplied) Stalk-eyed flies (family Diopsidae) are a model system for studying sexual selection due to the elongated and sexually dimorphic eye-stalks found in many species. These flies are of additional interest because their X chromosome is derived largely from an autosomal arm in other flies. To identify candidate genes required for development of dimorphic eyestalks and investigate how sex-biased expression arose on the novel X we compared gene expression between males and females using oligonucleotide microarrays and RNA from developing eyestalk tissue or adult heads in the dimorphic diopsid, Teleopsis dalmanni. more...

CGH reveals a neo-X chromosome and biased gene movement in stalk-eyed flies (genus Teleopsis)

(Submitter supplied) In this study, we utilized Comparative Genomic Hybridization (CGH), using probes designed from EST sequence, to identify genes located on the X chromosome of four species in the stalk-eyed fly genus Teleopsis. Analysis of log ratio values from the CGH microarrays for over 3400 genes produces a strongly bimodal distribution that clearly differentiates autosomal from X-linked genes for all four species. more...

Microarray analysis of stalk-eyed fly lines selected for divergent eyespan

(Submitter supplied) Eye-stalks of some flies exhibit extreme sexual dimorphism, but little is known about the genetic mechanisms producing variation in these ornamental traits. Therefore, we constructed an EST database of genes expressed in the developing eye-antennal imaginal disc of the highly dimorphic species, Teleopsis dalmanni, and used this set of genes to construct high density oligoarrays and compare patterns of gene expression between lines of flies selected for divergent eyespan. more...

Agilent-020839 Teleopsis dalmanni 579_FLS_slide_II (condensed version)

(Submitter supplied) Probes were based on EST sequences obtained from eye-imaginal disc tissue. A total of 3424 annotated genes along with 274 contigs with either large open reading frames (>500 bp) or relatively high representation in the EST database (>4 cDNA clones). In a previous study, we created 10 probes for each gene and used them to conduct comparative genomic hybridization for several Teleopsis species, including T. more...

Tdalmanni Agilent 44K Oligoarray Slide1

(Submitter supplied) Each 44K array contains 9 features for each of ~3600 genes that have been identified in an EST study of eye-antennal discs from late instar larvae or pupae of a diopsid stalk-eyed fly. cDNA clusters were included that had 1) significant homology to known genes in Drosophila or Anopheles, 2) an ORF larger than 300 bp (labeled ORF-) or 3) more than four associated cDNA clones (labeled X-). Three 60 bp oligonucleotide probes were designed for each gene based on the consensus sequence with the highest Blast hit or largest ORF within each cluster. more...
