CD16+ CD163+ monocytes traffic to sites of inflammation in NEC

(Submitter supplied) Using suspension (CyTOF) and imaging (IMC) mass cytometry coupled with single cell RNA sequencing (scRNAseq) on intestinal mucosa, we demonstrated severe inflammation in patients with NEC. scRNAseq revealed that the CD163+CD16+ monocytes/Mf transcribe a large number of inflammatory genes including TREM1, IL1a and IL1b, IL8 and calprotectin. Gene set enrichment analysis identified pathways involved in chemotaxis and migration to be enriched in the clusters containing CD163+CD16+ monocytes/Mf, confirming our observations that they traffic from the periphery.

Gene Expression in Transformed Lymphocytes Reveals Phenotype-Modifying Pathways in Cystic Fibrosis

(Submitter supplied) Heritable genetic variants modify cystic fibrosis (CF) clinical phenotypes, e.g., lung disease, age-of-onset of persistent Pseudomonas aeruginosa (P. aeruginosa), and meconium ileus (MI).  Previous genome wide association studies (GWAS) have begun to inform the genetic architecture of CF phenotypes.  Analyses of gene expression will complement GWAS, as demonstrated by analyses of gene expression in lymphoblastoid cell lines (LCLs) to identify disease-related pathophysiological processes for non-CF complex traits.  In this study, global gene expression was measured in RNA from LCLs from 754 CF patients and analyzed for association with lung disease severity, age-of-onset of persistent P. more...

Genome-wide analysis of the transcriptional response to porcine reproductive and respiratory syndrome virus infection at the maternal/fetal interface and in the fetus

(Submitter supplied) Porcine Reproductive and Respiratory Syndrome Virus (PRRSV) infection of 3rd trimester pregnant pigs can result in transmission of the virus to the fetus and ultimately death in utero or postnatally. Little is known about the immune response to infection at the maternal-fetal interface and in the fetus itself, or the molecular events behind virus transmission and disease progression in the fetus. To investigate these processes, RNA-sequencing of two tissues, uterine endothelium adjacent to the umbilical attachment site and fetal thymus, was performed 21 days post challenge on four groups of fetuses selected from a large PRRSV challenge experiment of pregnant gilts.

Gene expression of metamorphosis in fire ants

(Submitter supplied) Ants are among the most successful animals on earth, with societies of a complexity that rivals our own. These societies are characterized by reproductive division of labor between female queens that can live several years and lay thousands of eggs per day, workers that live only a few months and are sterile, and males that live only a few weeks and do not participate in colony tasks. These striking differences in lifespan and roles are echoed by extensive morphological and physiological divergence. more...
