Human PXR signaling enhances ethanol-induced liver injury in females

(Submitter supplied) Alcohol-associated liver disease (ALD) results from excessive alcohol consumption and is more severe in females. The prenane X receptor (PXR), a xenobiotic nuclear receptor essential for toxin defense, has been implicated in ALD, but the underlying mechanisms remain unclear. Due to species differences in ligand specificty, humanized PXR (hPXR) mouse models are necessary to investigate its role in the sexual dimorphism of ALD in humans. more...

Expression of a murine model of Rheumatoid Arthritis with or without Intermittent fasting

(Submitter supplied) Intermittent fasting (IF) reduces the inflammatory symptoms of Rheumatoid Arthritis (RA); however, its anti-inflammatory underlying molecular mechanisms are unknown. Aim: To investigate the effect of IF on joint inflammatory molecular mechanisms in collagen-induced arthritis (CIA) through DNA microarray and clinical and histopathological evaluations. Methods: CIA was induced in sixteen male DBA/1 mice divided into two groups, one received every other day IF for four weeks. more...

Transcriptome analysis of coding and ncRNA isoforms in monocytes and macrophages purified from bone marrow of xenotransplanted mice during leukemia progression.

(Submitter supplied) Immune cells of the myeloid lineage and CLL cells support each other during leukemia progression and dissemination. We have investigated the molecular interactions supporting this cell-cell interdependence with a special focus on ncRNAs and microRNAs, whose role in nonmalignant monocytes and macrophages is largely unknown. In the CLL xenograft model based on the engraftment of MEC1 human CLL cell line into Rag2-/-γc-/-, we analyzed the transcriptome of monocytes/macrophages isolated from the bone marrow during leukemia progression. more...

mRNA ADENOSINE METHYLASE promotes drought tolerance through N6-methyladenosine-dependent and independent impacts on mRNA regulation in Arabidopsis

(Submitter supplied) • Among many mRNA modifications, adenine methylation at the N6 position (N6-methyladenosine, m6A) is known to affect mRNA biology extensively. The influence of m6A has yet to be assessed under drought, one of the most impactful abiotic stresses. • We show that Arabidopsis thaliana (L.) Heynh. (Arabidopsis) plants lacking mRNA ADENOSINE METHYLASE (MTA) are drought sensitive. Subsequently, we comprehensively assess the impacts of MTA-dependent m6A changes during drought on mRNA abundance, stability, and translation in Arabidopsis. more...

129-derived passenger mutations confound phenotype of ACKR1-deficient mice

(Submitter supplied) A majority of genetically modified mice carry passenger mutations, originating from the 129-derived embryonic stem (ES) cells, near the targeted gene. Unintended retention of these mutations can introduce confounding phenotypes and affect conclusions. We show that Ackr1-/-129ES mice retained approximately 6 Mb of 129 ES cells-derived genomic material in chromosome 1 near Ackr1 in bone marrow derived monocytes (Mono), nucleated erythroblasts (NECs), and polymorphonucleated neutrophils (PMN), notably affecting the expression of PYHIN genes.

Effect of PRMT5 inhibitor on gene expression in MTAP null pancreatic cell lines

(Submitter supplied) One of the most robust synthetic lethal interactions observed in multiple functional genomic screens has been the dependency on PRMT5 in cancer cells with MTAP deletion. Here we report the discovery of the clinical stage MTA-cooperative PRMT5 inhibitor, AMG 193. AMG 193 preferentially binds PRMT5 in the presence of MTA and has potent biochemical and cellular activity in MTAP-deleted cells across multiple cancer lineages. more...

Transcriptional changes after controlled cortical impact in mice

(Submitter supplied) Many extracellular matrix (ECM) changes occur in the brain after traumatic brain injury. This work sought to understand the dynamics of ECM modifications after TBI by comparing RNA transcription between ipsilateral and contralateral brain regions. Mice underwent controlled cortical impact (with a 2mm depth) using a pneumatic impactor. Seven days later, brain tissue was harvested from the site of injury and from the corresponding contralateral cortex. more...

Expression Data from ST2 cells transfected with a Wnt1 expressing plasmid

(Submitter supplied) Wnt1 has major anabolic functions in bone metabolism. We investigated the effect of of Wnt1 on the mesenchymal cell line ST2 by transfecting it with a pLNCX-Wnt1 expression plasmid. We used microarray analysis to detail the global gene expression of ST2 cells 5 days after transfection with 1 µg pLNCX-Wnt1.

Postnatal Allergic Inhalation Induces Glial Inflammation in the Olfactory Bulb and Leads to Autism-Like Traits in Mice

(Submitter supplied) Autism spectrum disorder (ASD) is one of the most prevalent neurodevelopmental disorders. To clarify its detailed pathophysiology, we studied the association between neonatal allergic exposure and behavioral changes. Adult female C57BL/6J mice were immunized with adjuvant (alum) or ovalbumin emulsified by adjuvant. After immunization, mice were mated, and offspring were born in full-term. Postnatal mother and infant were simultaneously inhaled with allergen. more...

Experimental menopause in 3xTg-AD mice triggers metabolic and cognitive dysfunction aligned with Alzheimer's disease processes

(Submitter supplied) There is a compelling link between menopause and Alzheimer's disease (AD), with few established molecular mechanisms. A lack of experimental models limits validation of causal drivers of these interactions. Here, we characterize the accelerated ovarian failure (OF) model of menopause in the triple-transgenic AD (3xTg-AD) mouse. Ovotoxin, 4-vinylcyclohexene diepoxide accelerated follicular loss, ablating circulating progesterone. more...

Ablation of Max expression induces meiotic onset in sexually undifferentiated germ cells

(Submitter supplied) MAX (MYC Associated Factor X) is generally known as a mandatory partner for MYC transcription factor, which activates various genes involved in cell growth and metabolism. On the other hand, MAX, when interacting with MGA, forms the polycomb repressive complex (PRC) 1.6, one of the subtypes of PRC1, which directs the transcriptionally repressed chromatin state. Although physiological significance is not known at present, we have previously demonstrated that mouse embryonic stem cells (ESCs) bear a potential to onset meiosis, albeit not germ cells, and PRC1.6 prevent ESCs from their ectopic onset of meiosis (Suzuki et al., 2016, Nat. more...

Chronic stress alters hepatic metabolism and thermodynamic respiratory efficiency affecting epigenetics in C57BL/6 mice [MeDIP-seq]

(Submitter supplied) Chronic stress episodes increase metabolic disease risk even after recovery. We propose that persistent stress detrimentally impacts hepatic metabolic reprogramming, particularly mitochondrial function. In male C57BL/6 mice chronic variable stress (Cvs) reduced energy expenditure (EE) and body mass despite increased energy intake versus controls. This coincided with decreased glucose metabolism and increased lipid β-oxidation, correlating with EE. more...

Gene expression profile of hair follicle stem cells (HFSCs) isolated from wild type and homozygous Dab2 cKO (Dab2 Fl/Fl) mice at postnatal day 35

(Submitter supplied) Disabled 2 is an endocytic adaptor protein known to regulate various signaling pathways such Wnt , BMP, TGF and Ras/MAPK. In the present study, we observed that Dab2 is expressed in all the compartments of the hair follicle. K14 Cre specific homozygous Dab2 knockout resulted in delayed activation of HFSCs and delayed Ist hair follicle cycle initiation. At PD35, while WT progresses to anagen II, the Dab2 cKO mice still remains at telogen. more...
