Chitinase 1: A novel therapeutic target in metabolic dysfunction-associated steatohepatitis

(Submitter supplied) Macrophage profiling was performed using single-nuclei RNA sequencing and flow cytometry. Single-nuclei RNA sequencing demonstrated a notable increase in macrophages numbers, particularly of lipid-associated macrophages, in MASH mice.

ARNT2 Controls Prefrontal Somatostatin Interneurons Mediating Affective Empathy

(Submitter supplied) Empathy is crucial for our social lives, and its disruption is a prominent characteristic of various psychiatric conditions. However, the specific genes and neurobiological mechanisms underlying empathy deficits remain elusive. By combining forward genetic mapping with transcriptome analysis, we discovered that the Arnt2 gene encoding a basic-helix-loop-helix (bHLH)-PAS transcription factor is a key driver of significant alteration in observational fear, a basic form of affective empathy. more...

Cyclo His-Pro halts skeletal muscle degeneration and cardiomyopathy in murine myopathy models

(Submitter supplied) Among the inherited myopathies, a group of muscular disorders characterized by structural and metabolic impairments in skeletal muscle, Duchenne muscular dystrophy (DMD) stands out for its devastating progression. Caused by the absence of functional dystrophin, DMD pathogenesis is driven by the progressive degeneration of muscle fibers, resulting in the development of inflammation and fibrosis that ultimately affect the overall muscle biomechanics. more...

Hypoxia promotes proliferation and inhibits myogenesis in broiler satellite cells

(Submitter supplied) Breast muscle myopathies in broilers compromise meat quality and continue to plague the poultry industry. Broiler breast muscle myopathies are characterized by impaired satellite cells (SC)-mediated repair, and localized tissue hypoxia and dysregulation of oxygen homeostasis have been implicated as contributing factors. The present study was designed to test the hypothesis that hypoxia disrupts the behavior of SC essential for multinucleated myotube formation in vitro, and to determine the extent to which effects are reversed by restoration of oxygen tension. more...

PRMT1 is required for antibody affinity maturation by preventing premature B cell differentiation

(Submitter supplied) Germinal center (GC) B cells undergo cycles of somatic hypermutation and selection, thus increasing their antibody affinity before differentiating into plasma or memory cells. The mechanisms dictating the dynamics of GC B cells are incompletely understood. We show that ablating the Protein arginine methyltransferase 1 (PRMT1) in GC B cells reduces antibody affinity maturation by impairing GC dynamics, as shown by compromised GC expansion, accumulation of CXCR4- B cells, and increased memory B cell differentiation, partly, at the expense of plasma cell production. more...

RNA-sequencing of gonadal adipose tissue from Chow and HFD fed MBOAT7 Flox and ASKO mice

(Submitter supplied) Non-alcoholic fatty liver disease (NAFLD) is becoming increasingly common and is a leading cause of end stage liver diseases such as cirrhosis and hepatocellular carcinoma. The rise in NAFLD closely parallels the global epidemic of obesity and type 2 diabetes mellitus (T2DM), and there is a clear interrelationship between abnormal lipid metabolism, insulin resistance, and NAFLD progression. Several genetic loci have been identified as contributors to NAFLD progression, all of which are consistently linked to abnormal lipid metabolic processes in the liver. more...

Istradefylline (KW6002) protects from cisplatin-induced nephrotoxicity and peripheral neuropathy while preserving cisplatin anti-tumor effects [Kidney]

(Submitter supplied) Cisplatin is a potent chemotherapeutic drug, widely used in the treatment of various solid cancers. However, its clinical effectiveness is strongly limited by frequent severe adverse effects, such as nephrotoxicity and chemotherapy-induced peripheral neuropathy. Therefore, there is an urgent medical need to identify novel strategies that would limit cisplatin-induced toxicity. In the present study, we provide evidence that the FDA-approved adenosine A2A receptor antagonist istradefylline (KW6002) significantly protects from cisplatin-induced nephrotoxicity and neuropathic pain in experimental models of acute and sub-chronic cisplatin intoxication. more...

Enteric glial cells favor accumulation of anti-inflammatory macrophages during the resolution of muscularis inflammation

(Submitter supplied) Monocyte-derived macrophages (Mφs) are crucial regulators during muscularis inflammation. However, it is unclear which micro-environmental factors are responsible for monocyte recruitment and anti-inflammatory Mφ differentiation in this paradigm. Here, we investigate Mφ heterogeneity at different stages of muscularis inflammation and determine how environmental cues can attract and activate tissue-protective Mφs. more...

Genomic signature of Fanconi anemia DNA repair pathway deficiency in cancer

(Submitter supplied) Fanconi anemia (FA), a model syndrome of genome instability, is caused by a deficiency in DNA interstrand crosslink (ICL) repair resulting in chromosome breakage. The FA repair pathway protects against carcinogenic endogenous and exogenous aldehydes. Individuals with FA are hundreds to thousands-fold more likely to develop head and neck (HNSCC), esophageal and anogenital squamous cell carcinomas (SCCs). more...

BET proteins regulate gene expression in early kidney development

(Submitter supplied) In utero renal development is subject to maternal metabolic and environmental influences af-fecting long-term renal function and the risk to develop chronic kidney failure and cardiovascular disease. Epigenetic processes have been implicated in the orchestration of renal development and prenatal programming of nephron number. Bromodomain and extraterminal domain proteins act as histone acetylation reader molecules and promote gene transcription. more...

Transcriptomic profiles of splenic Gr-1+ cells from IDO-KO hosts and WT hosts after bone marrow transplantation

(Submitter supplied) Tryptophan-catabolizing enzyme indoleamine 2,3-dioxygenase 1 (IDO1) has also immunological function to suppress T cell activation in inflammatory circumstances, including graft-versus-host disease (GVHD), a fatal complication after allogeneic bone marrow transplantation (allo-BMT). Although the mononuclear cell expression of IDO1 has been associated with improved outcomes in GVHD, the underlying mechanisms remain unclear. more...

Multiomics profiling of HIV-infected cells

(Submitter supplied) The restructuring of chromatin architecture following lentiviral integration is not well elucidated. We jointly interrogate (HIV-distal & -local) chromatin organization (via Hi-C & ATAC-seq) and the RNA landscape around defined sites of proviral integration using HIV-inducible cellular models. We report chromatin interaction networks and nuclear ultrastructure around integrated HIV-1 are predominantly preserved, suggesting HIV integration does not induce large scale remodeling of cellular chromatin. more...

Recently, it has reported that the genetic ablation of Crif1 in a tissue-specific manner results in mitochondrial stress response (MSR) including mitochondrial unfolded protein response (UPRmt). more...
