Ventral hippocampal transcriptome analysis after acute corticosterone exposure.

(Submitter supplied) Aim: We investigated the effect of acute corticosterone exposure 3 hours after subcutaneous injection on the ventral hippocampus transcriptome. Methods: Adult male mice were used in the experiment. Mice were sacrificed 3 hours after corticosterone (3mg/kg) of vehicle injection and the ventral hippocampus was dissected and snap-frozen. Subsequently, total RNA was isolated and send for 100bp paired-end sequencing by BGI.

Hepatic transcriptome analysis after acute and chronic glucocorticoid exposure combined with androgen receptor antagonism.

(Submitter supplied) Aim: We investigated the effect of acute and chronic glucocorticoid exposure on the hepatic transcriptome and its dependency on active androgen signaling. Methods: Adult male mice were used in the experiments. For the chronic exposure, mice were implanted with CORT (20mg) or vehicle pellets and administered ENZA (40mg/kg/day) or solvent via oral gavage. Reproductive tract extracellular vesicles are sufficient to transmit intergenerational stress and program neurodevelopment

(Submitter supplied) We report reproductive tract extracellular vesicles (EVs) transmit information regarding stress in the paternal environment to sperm, ultimately altering fetal development. Using intracytoplasmic sperm injection, we found that sperm incubated with EVs collected from stress-treated epididymal epithelial cells produced offspring with altered neurodevelopment and adult stress reactivity. Proteomic and transcriptomic assessment of these EVs showed dramatic changes in protein and miRNA content long after stress treatment had ended.

Transcriptomic response of the arcuate nucleus to oral corticosterone treatment

(Submitter supplied) Glucocorticoids (GCs) are widely prescribed to treat inflammatory and autoimmune conditions. However, patients often develop adverse metabolic effects, including hyperphagia, obesity, and hyperglycemia.The arcuate nucleus has an established role in the central control of energy balance. In this study we examined the transcriptomic response of the arcuate nucleus to oral corticosterone treatment. Gene expression changes were measured after 2 days and 4 weeks of Cort treatment in ad libitum fed C57BL6/J mice.

Effect of Mineralocorticoid Receptor deletion on glucocorticoid signalling in the macropahge

(Submitter supplied) Inappropriate excess of the steroid hormone aldosterone, which is a mineralocorticoid receptor (MR) agonist, is associated with increased inflammation and risk of cardiovascular disease. MR antagonists are cardioprotective and antiinflammatory in vivo, and evidence suggests that they mediate these effects in part by aldosterone- independent mechanisms. We used affymetrix to characterize the effect of Mineralocorticoid Receptor deletion on macrophage transcriptional profile, and identify its requirement in normal glucocorticoid signalling.
