Identification of HTRA4 as a trasncritpional target of P63 in trophoblast

(Submitter supplied) The placenta plays a crucial role in pregnancy success. deltaNp63alpha (p63), a transcription factor from the TP53 family, is highly expressed in villous cytotrophoblasts (CTBs), the epithelial stem cells of the human placenta, and has been shown to be involved in CTB maintenance and differentiation. In this study, we set out to identify mechanism(s) of action of p63, by identifying its downstream targets. more...

Association of SNPs in the PAI1 Gene with Disease Recurrence and Clinical Outcome in Bladder Cancer

(Submitter supplied) Bladder cancer is one of the most common cancer types worldwide and is characterized by a high rate of recurrence. Previous studies have demonstrated that plasminogen activator inhibi-tor-1 (PAI1) plays an important role in bladder cancer development. The aim of our retrospective study was to assess the effects of PAI1 mutational status in bladder tumors. In this study, we evaluated the mutational status of PAI1 in a series of independent cohorts, comprised of a total of 660 subjects. more...

Coordinated regulation of protein synthesis and mitochondrial respiration by the molecular chaperone TRAP1 in cancer cells

(Submitter supplied) Here we show that TRAP1 directly binds translation elongation factors, both inside and outside mitochondria, and slows down translation. TRAP1 overexpression or silencing affects the synthesis of respiratory complex components. Inside mitochondria, TRAP1 binds the Complex III core component UQCRC2 and regulates Complex III activity. This decreases respiration rate upon basal condition but allows sustained oxidative phosphorylation when glucose is limiting, a condition in which TRAP1-UQCRC2 binding is lost. more...

Proteotranscriptomic decoding of the surface-endocytome in primary glioblastoma cells identifies potential target antigens in the hypoxic tumor niche [U87_mRNA_Norm_Hyp]

(Submitter supplied) Background: Antibody and cell-based immunotherapies, e.g., antibody-drug-conjugates and CAR-T cells, targeted at cell-surface proteins, currently revolutionize clinical oncology. However, target selection warrants a better understanding of the surface-endocytome and how it is modulated by the tumor microenvironment. Here, we unravel the surface-endocytome landscape and its remodeling by hypoxia in primary cultures from glioblastoma (GBM) patients that currently lack targeted therapies. more...

FBP1 overexpression in MDA-MB-231 breast cancer cells

(Submitter supplied) Fructose 1,6 bisphosphatase the key enzyme of gluconeogenesis has emerged as a potential metabolic tumor suppressor. In breast cancer distinct FBP1 expression levels are found depending on subtype. In MDA-MB-231 FBP1 expression is suppressed. To get more insight into potential functions of FBP1 we introduced a vector to reintroduce expression of FBP1. These experiments are part of a more comprehensive study on expression of FBP1 in breast cancer cells.

Gene expression profiles of endometrium from women across the menstrual cycle

(Submitter supplied) Natural variability in menstrual cycle length, coupled with rapid changes in endometrial gene expression, makes it difficult to accurately define and compare different stages of the endometrial cycle. We have developed and validated a novel method for precisely determining endometrial cycle stage based on global gene expression. Our ‘molecular staging model’ reveals significant and remarkably synchronized daily changes in expression for over 3,400 endometrial genes throughout the cycle, with the most dramatic changes occurring during the secretory phase.

Inflammatory markers and frailty in home-dwelling elderly, a cross-sectional study

(Submitter supplied) Low-grade, chronic inflammation during ageing (“inflammageing”) is suggested to be involved in the development of frailty in older age. However, studies on the association between frailty, using the frailty index definition, and inflammatory markers are limited. The aim of this study was to investigate the relationship between inflammatory markers and frailty index (FI) in older, home-dwelling adults. more...

HeberFERON distinctively targets Cell Cycle in the glioblastoma-derived cell line U-87MG

(Submitter supplied) Transcriptomic profile study comparing human glioblastoma derived cell line U-87MG treated with HeberFERON, alpha and gamma interferons. HeberFERON is a co-formulation of alpha2b and gamma interferons, based on their synergism, that have shown its clinical superiority over individual interferons in basal cell carcinomas. In Glioblastoma (GBM), HeberFERON has shown promising preclinical and clinical results. more...

Microarray analysis of lung cancer cells gene expression in overexpressing different semaphorin 5a domains

(Submitter supplied) Semaphorin 5A (SEMA5A) is one member of semaphorin family, which was identified as an axon guidance molecule in nervous systems. In our previous studies, SEMA5A was identified as a prognostic biomarker for lung cancer in nonsmoking women and acted as a tumor suppressor gene in inhibiting the proliferation and migration of lung cancer cells. Yet, the regulatory mechanism of SEMA5A was not clear. Therefore, the purpose of this study was to explore the roles of different domains of SEMA5A on the tumor suppressive effects in lung cancer cell lines.

C99R mutation in IRF4 drives a novel gain of function binding and gene upregulation in classical Hodgkin lymphoma [Illumina BeadChip]

(Submitter supplied) Disease-causing mutations in genes encoding transcription factors (TF) are frequent in hematopoietic malignancies and might affect TF-interactions with respective DNA-binding motifs. Here, we report the characterization of a recurrent somatic mutation c.295T>C in the IRF4 gene in classic Hodgkin lymphoma (cHL) leading to a C99R exchange. IRF4-C99R is located in the center of the IRF4 alpha3-recognition helix contacting the DNA 5´-GAAA-3´ IRF consensus sequence. more...

CTLA-4 pathway is instrumental in Giant Cell Arteritis

(Submitter supplied) BACKGROUND: Giant Cell Arteritis (GCA) causes severe inflammation of the aorta and its branches and is characterized by intense effector T cells infiltration. The roles that immune checkpoints play in pathogenesis of GCA are still unclear. Our aim was to study the immune checkpoints interplay in GCA. METHODS: First, we used VigiBase, the WHO international pharmacovigilance database, to evaluate the relationship between GCA occurrence and immune checkpoint inhibitors (ICI) treatments. more...

KLF10 modulates stem cell phenotypes of pancreatic adenocarcinoma by transcriptionally regulating notch receptors

(Submitter supplied) KrÜppel-like factor 10 (KLF10), deficient in two thirds of 105 resected pancreatic adenocarcinoma (PDAC) patients, was associated with rapid loco-regional recurrence and large tumor size. Additional KLF10 depletion in KC (LSL: KrasG12D; Pdx1-CRE) mice accelerated progression from pancreatic intraepithelial neoplasia to PDAC. In Panc-1 cells with KLF10 deficiency (Panc-1-pLKO-shKLF10), increased sphere formation, stem cell markers expression, and tumor growth were noted compared with those of control. more...
