CRISPR screen identifies loss of IFNγR signaling and downstream adhesion as a resistance mechanism to CAR T-cell cytotoxicity in solid tumors

(Submitter supplied) U87-CBG-GFP cells selected with blasticidin to stably express Cas9 were transduced with an MOI (< 1) of Brunello guide library (with puromycin resistance in the guide backbone). Cells were rested for 2 days then selected with puromycin antibiotic until the control, untransduced arm was killed (6-7 days). The guide-transduced cells were then pooled and replated in preparation for the screen. There were 12 arms to the screen, two control arms performed in technical duplicate and 8 sample arms from 6 normal donor CAR T-cells (2 were run in technical duplicate). more...

Transcriptome response of Gluconobacter oxydans against five typical aldehyde inhibitors

(Submitter supplied) Toxic inhibitory compounds from lignocellulose pretreatment are the major obstacle to achieve high bioconversion efficiency in biorefinery fermentations. This study shows a unique glucose oxidation catalysis of Gluconobacter oxydans with its gluconic acid productivity free of inhibitor disturbance. The microbial experimentations and the transcriptome analysis revealed that both the activity of the membrane-bound glucose dehydrogenase (mGDH) and the transcription level of the genes in periplasmic glucose oxidation respiratory chain of G. more...

Systems-level Analysis of Oxygen Exposure in Zymomonas mobilis: Implications for Isoprenoid Production

(Submitter supplied) Zymomonas mobilis is an aerotolerant anaerobe and prolific ethanologen with attractive characteristics for industrial bioproduct generation. However, there is currently insufficient knowledge of the impact that environmental factors have on flux through industrially relevant biosynthetic pathways. Here, we examine the effect of oxygen exposure on metabolism and gene expression in Z. mobilis by combining targeted metabolomics, mRNA sequencing, and shotgun proteomics. more...

Heavy metal resistance in Cupriavidus metallidurans CH34 is governed by an intricate transcriptional network

(Submitter supplied) Background: The high number of heavy metal resistance genes in the soil bacterium Cupriavidus metallidurans CH34 makes it an interesting model organism to study microbial responses to heavy metals. Results: In this study the transcriptional response of this bacterium was measured after challenging it to a wide range of sub-lethal concentrations of various essential or toxic metals. Considering the global transcriptional responses for each challenge as well as by identifying the overlap in upregulated genes between different metal responses, the sixteen metals could be clustered in three different groups. more...

Carbon nutrition of pathogenic and commensal E. coli

(Submitter supplied) Transcriptome profiling of E. coli MG1655, E. coli EDL933, E. coli ECOR26, and E. coli Nissle 1917 grown on mouse cecal mucus as carbon source The carbon sources that support growth of pathogenic E. coli O157:H7 in the mammalian intestine have not previously been investigated. In vivo, pathogenic E. coli EDL933 primarily grows as dispersed single cells within the mucus layer that overlies the mouse cecal epithelium. more...
