Immune checkpoint inhibitor monotherapy is sufficient to promote microenvironmental normalization via Type I interferon pathway in PD-L1+ expressing head and neck cancer

(Submitter supplied) Immune checkpoint blockers (ICBs) targeting programmed death 1(PD-1) are considered effective first-line therapy against PD-1 ligand (PD-L1)-expressing head and neck squamous cell carcinoma (HNSCC). However, associated changes in tumor microenvironment (TME) and mechanisms remain elusive. Oral tumors in C57/BL6 mice were induced by administering 7,12-dimethylbenzanthracene into the buccal mucosa. Single-cell suspension was isolated from tumor tissue; proliferating cells were injected subcutaneously into the left flank of mice to establish Ajou Oral Cancer (AOC) cell lines. more...

Intestinal myofibroblast vs skin fibroblast

(Submitter supplied) Pericryptal myofibroblasts in the colon and rectum play an important role in regulating the normal colorectal stem cell niche and facilitating tumour progression. Myofibroblasts have previously mostly been distinguished from normal fibroblasts only by the expression of α smooth muscle actin (αSMA). We now identify AOC3, a surface monoamine oxidase, as a new marker of myofibroblasts by showing that it is the target protein of the myofibroblast reacting monoclonal antibody (mAb), PR2D3. more...

Microarray analysis of med16, med8, elp2, and wild type (Col-0) infected with the necrotrophic fungal pathogen Sclerotinia sclerotiorum

(Submitter supplied) The goal of the microarray experiment was to identify defense genes that were differentially expressed in the Arabidopsis mutants med16, med8, elp2, wild type in response to infection of the necrotrophic fungal pathogen Sclerotinia sclerotiorum. Results indicated that, compared with the wild type, the jasmonic acid (JA) biosynthesis genes LOX3, AOC3, and OPR3, ethylene (ET) biosynthesis genes ACS2 and ACS8, as well as salicylic acid (SA) biosynthesis genes ICS1 and EDS5 were up-regulated in med16 in local tissues at 1 day post-inoculation (dpi) and/or systemic tissues at 4 dpi. more...
