Description

This variant has been previously reported as a hemizygous change in patients with X-linked adrenoleukodystrophy (PMID: 11748843, 33920672; The ALD Variant Database https://adrenoleukodystrophy.info). Missense variation is an established mechanism of disease for ABCD1-related disorders (PMID: 11748843). The c.1903G>A (p.Val635Met) variant is absent from the gnomAD population database and thus is presumed to be rare. The c.1903G>A (p.Val635Met) variant affects a highly conserved amino acid and is predicted by multiple in silico tools to have a deleterious effect on protein function. Based on the available evidence, the c.1903G>A (p.Val635Met) variant is classified as Likely Pathogenic.

Description

In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt ABCD1 protein function. This missense change has been observed in individual(s) with features of X-linked adrenoleukodystrophy (PMID: 11748843; Invitae). The frequency data for this variant in the population databases is considered unreliable, as metrics indicate poor data quality at this position in the gnomAD database. This sequence change replaces valine, which is neutral and non-polar, with methionine, which is neutral and non-polar, at codon 635 of the ABCD1 protein (p.Val635Met).
