Description

This sequence change replaces serine, which is neutral and polar, with leucine, which is neutral and non-polar, at codon 187 of the FH protein (p.Ser187Leu). For these reasons, this variant has been classified as Pathogenic. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt FH protein function. ClinVar contains an entry for this variant (Variation ID: 393567). This variant is also known as C431T and S144L. This missense change has been observed in individual(s) with hereditary leiomyomatosis and renal cell cancer (PMID: 12772087; Invitae). This variant is present in population databases (rs398123166, gnomAD 0.004%).
