Description

The c.8676delA pathogenic mutation, located in coding exon 20 of the BRCA2 gene, results from a deletion of one nucleotide at nucleotide position 8676, causing a translational frameshift with a predicted alternate stop codon (p.R2892Sfs*17). This alteration has been reported in one French Canadian family at high risk of breast and/or ovarian cancer (Simard J et al. J Med Genet, 2007 Feb;44:107-21). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
