Description

This sequence change creates a premature translational stop signal (p.Val598Serfs*4) in the MYBPC3 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in MYBPC3 are known to be pathogenic (PMID: 19574547). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with MYBPC3-related conditions. ClinVar contains an entry for this variant (Variation ID: 181074). For these reasons, this variant has been classified as Pathogenic.

Description

The p.Val598fs variant in MYBPC3 has not been previously reported in the literature, but has been reported in ClinVar (Variation ID 181074). It was absent from large population studies. This variant is predicted to cause a frameshift, which alters the protein’s amino acid sequence beginning at position 598 and leads to a premature termination codon 4 amino acids downstream. This alteration is then predicted to lead to a truncated or absent protein. Heterozygous loss of function of the MYBPC3 gene is an established disease mechanism in HCM. In summary, the p.Val598fs variant meets criteria to be classified as pathogenic for HCM in an autosomal dominant manner based upon its predicted impact to the protein and absence from controls.
