Description

The c.1711+4A>T intronic variant results from an A to T substitution 4 nucleotides after coding exon 11 in the CDH1 gene. This nucleotide position is well conserved on limited sequence alignment. Using the BDGP and ESEfinder splice site prediction tools, this alteration is predicted to weaken the efficiency of the native splice donor site; however, direct evidence is unavailable. A similar alteration affecting this donor splice site (c.1711+5A>T or IVS11+5G>A) has been reported in an individual diagnosed with diffuse gastric cancer at age 48 who had family history of two relatives with confirmed diffuse gastric cancer and five relatives with breast cancer, one confirmed as lobular, and RT-PCR analyses indicated that this alteration led to exon 11 skipping (Brooks-Wilson AR et al. J Med Genet. 2004 Jul;41(7):508-17). Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
