Description

The p.T515S variant (also known as c.1544C>G), located in coding exon 14 of the FANCC gene, results from a C to G substitution at nucleotide position 1544. The threonine at codon 515 is replaced by serine, an amino acid with similar properties. In one study, this alteration was detected in 1/1441 breast cancer cases and 0/464 healthy controls (Thompson ER et al. PLoS Genet, 2012 Sep;8:e1002894). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
