Description

This variant has been observed in individual(s) with autosomal dominant Charcot-Marie-Tooth disease (PMID: 26392352, Invitae). In at least one individual the variant was observed to be de novo. ClinVar contains an entry for this variant (Variation ID: 653992). This variant is not present in population databases (ExAC no frequency). This sequence change replaces isoleucine with phenylalanine at codon 88 of the MFN2 protein (p.Ile88Phe). The isoleucine residue is highly conserved and there is a small physicochemical difference between isoleucine and phenylalanine. For these reasons, this variant has been classified as Pathogenic. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C15").
