Description

The COL11A2 c.539G>A; p.Arg180His variant (rs142462583), to our knowledge, is not reported in the medical literature but is reported in ClinVar (Variation ID: 439497). This variant is found in the general population with an overall allele frequency of 0.0090% (25/277948 alleles) in the Genome Aggregation Database. The arginine at codon 180 is weakly conserved, and computational analyses predict that this variant is neutral (REVEL: 0.017). Due to limited information, the clinical significance of this variant is uncertain at this time.

Description

This sequence change replaces arginine, which is basic and polar, with histidine, which is basic and polar, at codon 180 of the COL11A2 protein (p.Arg180His). This variant is present in population databases (rs142462583, gnomAD 0.01%). This variant has not been reported in the literature in individuals affected with COL11A2-related conditions. ClinVar contains an entry for this variant (Variation ID: 439497). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt COL11A2 protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
