Description

This sequence change inserts 1 nucleotide in exon 33 of the FBN1 mRNA (c.3973dupG), causing a frameshift at codon 1325. This creates a premature translational stop signal (p.Glu1325Glyfs*3) and is expected to result in an absent or disrupted protein product. While this particular variant has not been reported in the literature, loss-of-function variants in FBN1 are known to be pathogenic (PMID: 17657824, 19293843). For these reasons, this variant has been classified as Pathogenic.
