Model of a conserved pathway for regulating nuclear transport of transcription factors in Drosophila and mammals. (A) In Drosophila, the Toll protein binds the signal from the Spätzle protein and activates the kinase region of the Pelle protein. The Pelle protein phosphorylates Cactus and Dorsal, causing the two proteins to separate from each other. The Dorsal protein can then enter the nucleus and regulate the transcription of ventrally specific genes. (B) In mammalian lymphocytes, the IL-1 receptor can cause the phosphorylation of IκB (through the protein IRAK kinase). This enables the NF-κB protein to enter the nucleus and effect the transcription of several lymphocyte-specific genes involved in the inflammatory response. The components of the mammalian Toll-like receptor signaling pathway that culminates in the activation of NFκB have direct parallels in the components of the signaling pathway from the Toll receptor of Drosophila. The intracellular domain of the Toll-like receptors interacts with a homologous domain in the adaptor protein MyD88. A similar interaction occurs between the intracellular domain of Toll and dMyD88. The next step in both signaling pathways occurs via the interaction of death domains, between MyD88 and IRAK in the mammalian cells and between dMyD88 and Pelle in Drosophila. Both IRAK and Pelle are serine kinases. At this point the mammalian signaling pathway passes through an adaptor, TRAF6, which is activated by IRAK and in turn activates IKK. IKK phosphorylates the inhibitor of NFκB, IκB, targeting it for degradation and releasing the active dimeric transcription factor, NFκB. In Drosophila, homologues of MyD88, TRAF6, and of a kinase that is homologous to IKK that phosphorylates the Drosophila IκB homologue Cactus are found. Moreover, the terminal parts of the pathway are also homologous between Drosophila and mammals; phosphorylation of Cactus initiates its degradation and the release of the Dif/Relish dimer, which is a transcription factor and homologue of NFκB.

Spatially restricted activation of Toll establishes ventral cell fate in the early embryo. A) The positional information initiated in the oocyte is carried over to the fertilized egg. Initiated by early acting components such as Pipe in the oocyte, the serine protease cascade in the early embryo activates Spätzle by proteolytic processing. The mature Spätzle probably interacts directly with Toll. The signaling pathway causes degradation of Cactus, and allows Dorsal to enter the nuclei. B) Because the early step of activation occurs in ventral regions of the oocyte, the Toll pathway is only active in ventral regions of the embryo. The diffusible components of the pathway activate Toll gradually along the dorsal-ventral axis, leading to the formation of a nuclear gradient of Dorsal protein, with the highest level in ventral nuclei.
