Title Expression data from T-cell Acute Lymphoblastic Leukemia (T-ALL) patient-derived xenograft treated with anti-CD3 mAb
Organism Homo sapiens
Experiment type Expression profiling by high throughput sequencing
Summary T-cell acute lymphoblastic leukemia is a hematological malignancy treated by chemotherapy, with a poor prognosis. We have demonstrated that activation of the T cell receptor (TCR) by anti-CD3 antibodies has a potent anti-leukemic effect in patient-derived xenograft models (PDX) of T-ALL. Therefore, with this experiment, we aim to identify the molecular mechanisms underlying the anti-leukemic properties of the OKT3 anti-CD3 mAb in T-ALL PDX.

Overall design T-ALL PDX were treated with the isotype control antibody IgG2a or OKT3 mAb. After 6 hours of treatment, T-ALL cells were extracted and isolated from mouse cells by FACS sorting (hCD45+, hCD7+) for RNA collection. RNA-sequencing data were used to compare gene expression profiles upon OKT3 mAb treatment.
