Title Next Generation Sequencing Quantitative Analysis of altered expression of genes in lncRNA LNMAT1 knockdown bladder cancer cells

Summary Bladder cancer is one of the most common genitourinary malignancies worldwide, with approximately 429,800 new cases and 165,100 deaths annually in the world. LNMAT1 is reported to play diverse roles in the development and progression of human cancer. However, its function and underlying mechanism in bladder cancer remains unclear. The goal of this study is to identify the target genes of LNMAT1 in bladder cancer. Our results indicate that the genes regulated by LNMAT1 associated in a variety of biological functions, such as proliferation and metastasis.
 
Overall design To investigate the functional roles of LNMAT1 in bladder cancer, the UM-UC-3 and 5637 bladder cancer cell lines were established to stably downregulate LNMAT1 expression. The RNA of the transfected cells was extracted and hybridized on illumina Hiseq 2000. We indentified the putative target genes of LNMAT1 in both LNMAT1 shRNAs by comparing the different expressed genes among NC, sh-LNMAT1-1 and sh-LNMAT1-2. We further validated the data of next generation sequencing in UM-UC-3 and 5637 bladder cancer cells by qRT-PCR and western bloting.
