Results demonstrate that most mesopontine cholinergic neurons may transiently express VGLUT2, but that a large majority of parabigeminal nucleus neurons retain VGLUT2 expression throughout adulthood, and support a growing body of literature indicating that distinct cholinergic populations have differing potential for GABA or glutamate co-release.

Specifically, we show that mice lacking SRPX2 show a specific reduction in excitatory VGlut2 synapses in the cerebral cortex, while VGlut1 and inhibitory synapses were largely unaffected. SRPX2 KO mice also exhibit an abnormal ultrasonic vocalization ontogenetic profile in neonatal pups, and reduced preference for social novelty.

Study concludes that a variable number but functionally significant percentage of Intrinsically photosensitive retinal ganglion cells (ipRGCs) in two vGlut2 knockout mouse lines continue to release glutamate. Thus, the residual suprachiasmatic Nucleus -mediated light responses in these knockout mouse lines cannot be attributed solely to ipRGC pituitary adenylate cyclase-activating polypeptide release.

Vglut2 conditional knockout mice showed less interest in fatty rewards, decreased sugar consumption and strong rearing behavior, altered dopaminergic and peptidergic activity in the striatum. Significant cell loss and disorganization of the subthalamic nucleus structure was identified, which likely accounts for the observed alterations.
