Study: Effect of cpc-1 deletion in Neurospora crassa on transcriptome under different culture conditions

The high throughput mRNA-seq technology was used to measure the effect of cpc-1 deletion on transcriptome change of Neurospora crassa grown under different culture conditions using standard Illumina protocols. Overall design: The transcription profiles of wild-type (WT) strain FGSC4200 and cpc-1 deletion mutant (cpc-1?) of Neurospora crassa were generated by deep sequencing using Illumina sequencing platform. The WT and cpc-1? strains were cultured in 2% glucose medium (1?×?Vogel's, 2% glucose), 0.1% glucose medium (1?×?Vogel's, 0.1% glucose and 0.17% arginine) or 2% glucose medium with 5 mM 3-AT. A 5' epitope-tagged versions of cpc-1(WT) ORF under the control of the ccg-1 promoter and the ccg-1 5' untranslated region (5' UTR) was expressed in cpc-1? strain. This strain (cpc-1KO+cpc-1(WT)) was cultured in 2% glucose medium with 5 mM 3-AT.
