Study: Gut microbiota regulates tumor via circRNA/miRNA networks IV
show Abstracthide Abstract
Increasing evidence indicates that gut microbiota plays an important role in cancer progression. We have employed RNA-seq or microarray for genome including mRNA, microRNA or circRNA profiling in an gut microbiota -dependent manner, as a discovery platform to identify target genes with the potential to involve in tumor regulation. The deep sequencing analysis reveals regulatory functions of microbiota-mediated circular RNA (circRNA)/microRNA networks that may contribute to cancer progression. Overall design: Gut microbiota regulated mRNAs expression in tumor were measured in tumor samples of xenograft cancer model including germ-free mice with or without fecal microbiota transplantation (FMT) from specific pathogen free (SPF) mice or Probiotics (such as Bifidobacterium). Gut microbiota regulated no-coding RNAs (including circRNA and microRNA) expression in mice blood samples were measured in specific pathogen free (SPF) mice with or without broad-spectrum antibiotics (ABX) treatment, with or without fecal microbiota transplantation (FMT) from specific pathogen free (SPF) mice or Probiotics (such as Bifidobacterium, EcN) into germ-free mice.

Organism: Mus musculus
Library:
Instrument: NextSeq 500
Strategy: miRNA-Seq
Source: TRANSCRIPTOMIC
Selection: size fractionation
Layout: SINGLE
Construction protocol: RNA was harvested using Trizol reagent. Total RNA of each sample was used to prepare the miRNA sequencing library, which included the following steps:1) 3'-adaptor ligation;2) 5'-adaptor ligation3) cDNA synthesis;4) PCR amplification; 5) size selection of ~135-155 bp PCR amplified fragments (corresponding to ~15-35 nt small RNAs).
