High-throughput sequencing of plasma microRNA in elderly men fed a high protein diet in relation to the PBMC transcriptome: A 10 week randomized controlled trial Overall design: Thirty one healthy men older than age 70 were randomized to consume either the recommended dietary allowance (RDA; 0.8 g/kg/day) or twice the RDA (2xRDA; 1.6g/kg/day) of protein for 10 weeks.

Construction protocol: Plasma/Serum RNA Purification Kit using Norgen Biotek (Cat#55000) a standard library preparation workflow using Norgen's Small RNA Library Prep Kit for Illumina (Cat# 63600). Six microliter of each RNA sample was used as input. The workflow includes the following steps in sequence - 3' adapter ligation, 5' adapter ligation, reverse transcription/cDNA synthesis and then the indexing PCR. The final library is then subjected to column clean-up before resolving on a 6% TBE PAGE gel for size selection. The target band (~141-144 bp) is then excised from PAGE gel and purified
