Analysis of the contribution of the post-transcriptional regulator CsrA to translation during exponential growth in Escherichia coli Overall design: 4 biological replicates of ribosome protected fragments and paired RNA samples from wild type and csrA:: mutant strains in mLPM and LB were analyzed
Sample: ST_30: RNA csrA:: 2 mLPM

Layout: SINGLE
Construction protocol: Cell pellets were thawed and lysed with bead beating. Total RNA was extracted with miRNeasy RNA Extraction Kit (Qiagen). rRNA was subtracted with the Ribozero Gram Negative Kit (Illumina). RNA was then fragmented with heat and alkalinity and size selected between 20 and 40 bases with denaturing PAGE. RNA samples were ligated to a 3' adaptor and reverse transcribed. The cDNA was circularized to add a 5' adaptor then PCR amplified to add sample specific barcodes.
