Study: Transcriptome profile of ATP7B KO cells and WT cells treated with Copper (Cu) to study Autophagy as a pro-survival mechanism in Wilson disease
show Abstracthide Abstract
To understand whether hepatic cells use autophagy to defend against Cu toxicity in WD, we employed ATP7B knockout (ATP7B-KO) HepG2 cell line, by Quant-seq experiments. Transcriptome analysis of the of ATP7B KO cells and WT cells treated with Copper (Cu) compared with untreated cells. Overall design: total RNA was extracted from ATP7B KO cells and WT cells treated with Copper (Cu); RNA extracted from untreated cells was used as control.
Sample: Hep G2 wt cells untreated, biological rep3

Organism: Homo sapiens
Library:
Instrument: NextSeq 500
Strategy: RNA-Seq
Source: TRANSCRIPTOMIC
Selection: cDNA
Layout: SINGLE
Construction protocol: Extraction of total RNA was performed according to the manufacturer's instructions (Qiagen). Total RNA (100 ng) from each sample was prepared using QuantSeq 3'mRNA-Seq Library prep kit (Lexogen, Vienna, Austria) according to manufacturer's instructions The amplified fragmented cDNA of 300 bp in size were sequenced in single-end mode using the NextSeq500 (Illumina) with a read length of 75 bp
