To investigate a potential therapeutic effect of ABTAA on NV-AMD, we employed a mouse model of laser-induced CNV, that mimics human NV-AMD. A laser photocoagulation on mouse retinas was performed to rupture RPE and Bruch's membrane, and then the effects of ABTAA were compared with those of VEGF-Trap. Overall design: RPE-choroidal complex was obtained from enucleated eyes and flat-mounted on polyethylene naphthalate membrane (Carl Zeiss). CNV and surrounding tissues (a two-fold larger diameter than CNV itself), including RPE and choroid, were collected using laser capture microdissection (PALM MicroBeam, Carl Zeiss) and subjected to RNA isolation.

RNA libraries were prepared for sequencing using LEXOGEN Quant-Seq Library Prep Kit (Cat#001.24) standard protocols.
