show Abstracthide AbstractTo investigate a potential therapeutic effect of ABTAA on NV-AMD, we employed a mouse model of laser-induced CNV, that mimics human NV-AMD. A laser photocoagulation on mouse retinas was performed to rupture RPE and Bruch's membrane, and then the effects of ABTAA were compared with those of VEGF-Trap. Overall design: RPE-choroidal complex was obtained from enucleated eyes and flat-mounted on polyethylene naphthalate membrane (Carl Zeiss). CNV and surrounding tissues (a two-fold larger diameter than CNV itself), including RPE and choroid, were collected using laser capture microdissection (PALM MicroBeam, Carl Zeiss) and subjected to RNA isolation.
