show Abstracthide AbstractWe used male Sprague Dawley rats as experimental animals to establish a rat model of ASMVT (Acute Superior Mesenteric Venous Throwbosis) using the method described in a previous study (PMID: 34517186).Total RNA was extracted from three tissues each group. The RNA amount and purity of each sample was quantified using NanoDrop ND-1000 (NanoDrop, Wilmington, DE, USA). The RNA integrity was assessed by Agilent 2100 with RIN number >7.0. Small RNA sequencing library was prepared by TruSeq Small RNA Sample Prep Kits (Illumina, San Diego, USA). Next, deep sequencing was performed with an Illumina HiSeq2500 following the vendor's recommended protocol. The differentially expressed miRNAs were selected with statistical significance (p value < 0.05) by using Student t test. To predict the genes targeted by most aboundant miRNAs, two computational target prediction algorithms (TargetScan, v5.0 and Miranda, v3.3a) were used to identify miRNA binding sites. Finally, the data predicted by both algorithms were combined and the overlaps were calculated. The GO terms and KEGG Pathway of these most aboundant miRNAs, miRNA targets were also annotated. RNA-Seq analysis was performed by Hangzhou Lianchuan Biotechnology Co., Ltd (Hangzhou, China).
